摘要
该研究用5.5 Gy的γ射线照射大鼠,用Cocktail探针法结合液质联用、q PCR和Western blot法检测P450各个同工酶的酶活性、mRNA和蛋白表达来评价人参总皂苷对γ射线照射后大鼠肝P450酶调节作用。结果发现,辐射组CYP1A2,2B1,2E1,3A4与空白组相比,表达增加;人参总皂苷组CYP1A2,2B1,2E1,3A4与辐射组相比,表达下降。表明人参总皂苷对辐射诱导大鼠P450酶上调起到反向干预作用。
Effect of ginsenoside total saponin( GTS) on the regulation of P450 of livers of rats after γ-ray irradiation was studied.Rats were irradiated by the ^60Coγ-ray for one-time dose of 5. 5 Gy,dose rate of 117. 1-119. 2 c Gy. The cocktail probe,q PCR and Western blot were used to detect expression of enzymatic activites,mRNA andprotein of rats. Contrasted withblank group,expression of CYP1A2,2B1,2E1,3A4 of irradiation group showed a up-regulated( P〈0. 05). Contrasted with irradiation group,exprression of CYP1A2,2B1,2E1,3A4 of GTS group showed a downward trend. GTS had negative agonistic action against expression of P450 of rats by irradiatied.
