摘要
【目的】明确新疆部分地区籽瓜细菌性果斑病发生情况,分析不同分离物16S^23S rRNA的ITS区序列,确定不同分离物该基因区遗传变异情况。【方法】采集并分离来自阜康、额敏、沙湾等地的籽瓜以及五家渠甜瓜的病原细菌,应用细菌ITS区通用引物对各分离物16S^23S rRNA的ITS区序列进行扩增测序,并在GENEBANK数据库中作序列比对确定所分离病原种类,同时分析该ITS区序列差异。【结果】从四个地区籽瓜或甜瓜病组织上分离获得分离物均为噬酸菌属燕麦种西瓜亚种(Acidovorax avenae subsp.Citrulli),研究获得的7个果斑病菌分离物与国内外已报道7个分离物间ITS区序列同源性达99.5%~100%,该区遗传序列间并无显著差异。【结论】新疆阜康、额敏、沙湾等地籽瓜均有细菌性果斑病发生,参与比对的细菌性果斑菌分离物16S^23S rRNA的ITS区基因序列高度保守,该基因区遗传变异较小。
【Objective】This project aims to understand whether bacterial fruit blotch of watermelon has been infected edible seed watermelon in Xinjiang and understand whether genetic variation exists in the ITS region among different isolates by analyzing the ITS region of 16S- 23 S rRNA among different isolates.【Method】Pathogenic bacteria from edible seed watermelon were collected and isolated in Fukang,Eming,Shawan and melon in Wujiaqu. The 16S- 23 S rRNA ITS region genes of these isolates were amplified with universal primers L1 / L2. The PCR products were sequenced and their sequences were compared in Genebank,then pathogenic species were determined and the difference of ITS region gene among different isolates were analyzed.【Result】The result indicated that isolates isolated from edible seed watermelon and melon in four areas were Acidovorax avenae subsp. Citrulli. The sequence homology between seven isolates and other seven isolates reported in some literatures was 99. 5- 100 percent. There's no significant difference in the ITS region gene of these isolates. 【Conclusion】The bacterial fruit blotch was infected edible seed watermelon in Fukang,Eming,Shawan,but the ITS region of 16S- 23 S rRNA among different isolates in this research were highly conserved regions,and there's no significant difference in this gene regions.
