摘要
目的:筛选出朝天罐根抗炎活性部位,并研究其作用机制。方法:利用系统溶剂法对朝天罐根醇提取物进行萃取分离得到3个部分:乙酸乙酯部位、正丁醇部位及水液部位。采用二甲苯诱导小鼠耳廓肿胀、角叉菜胶致小鼠足趾肿胀实验综合评价不同提取部位的抗炎活性。采用RT-PCR法测定了乙酸乙酯部位对小鼠致炎耳组织表达的影响。结果:与空白对照组比较,朝天罐乙酸乙酯部位和正丁醇部位可显著降低小鼠耳廓肿胀和足肿胀度(P<0.05);乙酸乙酯部位可使致炎小鼠耳组织中肿瘤坏死因子(tumor necrosis factor-α,TNF-α)mRNA表达水平明显降低。结论:朝天罐乙酸乙酯部位和正丁醇部位为其抗炎活性部位,其作用机制为有效降低TNF-αmRNA的表达。
Objective:To investigate the anti-inflammatory fraction and mechanism of Radix Osbeckia opipara.Methods:Three polar fractions(ethyl acetate extracted fraction,n-butyl alcohol extracted fraction and water solution fraction)were prepared by extraction with organic solvents.The anti-inflammatory effects were evaluated in mice by using dimethyl benzene to induce auricle swelling,and carrageenan to induce foot swelling.Expression of mRNA of TNF-αwas detected by RT-PCR in ethyl acetate extracted fraction.Results:Ethyl acetate extracted fraction and n-butyl alcohol extracted fraction of Raadix Osbeckia opipara significantly reduce swelling of auricle and foot in mice(P〈0.05)compared with control group.Ethyl acetate extracted fraction reduced expression of TNF-α.Conclusion:The Ethyl acetate extracted fraction and nbutyl alcohol extracted fraction are effective anti-inflammatory fractions of Radix Osbeckia opipara.The anti-inflammatory mechanism is to reduce expression of TNF-α.
