摘要
目的观察瑞舒伐他汀对自发性高血压大鼠(SHR)主动脉血管重构和血管反应性的影响。方法雄性Wistar-Kyoto大鼠(WKY)为正常血压对照组(W组);SHR随机分为瑞舒伐他汀6.25 mg/(kg·d)组(R1组)、瑞舒伐他汀12.5 mg/(kg·d)组(R2组)、瑞舒伐他汀25 mg/(kg·d)组(R3组)、SHR对照组(S组)。瑞舒伐他汀溶于蒸馏水中灌胃,对照组给予等量蒸馏水灌胃,给药前后尾动脉测大鼠收缩压。8周给药结束后,取主动脉,一部分做HE染色观察血管重构,一部分用离体血管灌流法观察主动脉对不同药物的反应性。结果与W组比较,S组尾动脉收缩压显著增加,主动脉内径(L)显著减小,而中膜厚度(M)、中膜厚度与内径比值(M/L)显著增高。SHR主动脉对KCl(20 mmol/L^120 mmol/L)和苯肾上腺素(Phe,10-8mol/L^10-5mol/L)的收缩反应较W组显著增加;对乙酰胆碱(Ach,10-10mol/L^10-5mol/L)的舒张反应降低,但对硝普钠(SNP,10-10mol/L^10-4mol/L)的舒张反应差异无统计学意义。瑞舒伐他汀可使SHR收缩压的升高幅度降低,增加主动脉内径,降低中膜厚度,降低中膜厚度与内径比值(M/L);同时瑞舒伐他汀可以降低SHR主动脉对KCl和Phe的收缩反应,增强SHR主动脉对Ach的舒张反应,但对SNP的舒张反应无显著性影响。结论瑞舒伐他汀有延缓SHR收缩压升高的趋势,可以改善主动脉重构;增强SHR主动脉内皮依赖性舒张反应,降低其收缩反应,有效保护SHR血管内皮功能。
Objective To observe the effects of rosuvastatin on remodel and vascular reaction of aorta from spontaneously hypertension rats( SHR). Methods SHR and Wister-Kyoto( WKY) rats were randomly divided into: WKY control group,SHR treated with rosuvastatin 6. 25 mg /( kg·d),SHR treated with rosuvastatin 12. 5 mg /( kg·d); rosuvastatin 25 mg /( kg·d),SHR control group. Therapy continued for 8weeks. Tail systolic blood pressure was weekly measured before and after the study. At the end of this study,aorta was moved rapidly. A part of aorta was fixed with 4% paraformaldehyde to observe the vascular remodeling by HE stain. The vascular reaction of aorta was measured in vitro. Results Compared with WKY,the systolic blood pressure was higher,and vessel lumen diameter( L) decreased,while media thickness( M) and M/L increased in SHR. The contractions to phenylephrine( Phe),KCl in aorta were increased in SHR than in WKY. The relaxation to Ach decreased in SHR,but there was no difference of relaxation to SNP. Treatment with reosuvastatin induced a reduction in systolic blood pressure; increasing in L and decrease in M and M / L. Reosuvastatin reduced the contraction of aorta to Phe and KCl,increased the relaxation of aorta to Ach,but had no effect on the relaxation to SNP. Conclusion Reosuvastatin can retard the remodeling of aorta,improve the vascular reaction to different vasomotors,resume the balance of relaxation and contraction,protect endothelial function in SHR.
