摘要
目的探讨中药防风对感染后肠易激综合征(PI-IBS)大鼠模型肠道菌群及丝氨酸蛋白酶信号的影响。方法采用弯曲杆菌灌胃法复制PI-IBS大鼠模型,评价防风止痛、止泻的效应及对肠道菌群、粪便丝氨酸蛋白酶活性,结肠蛋白酶激活受体2(PAR-2)mRNA及紧密连接ZO-1蛋白表达的影响。结果防风可升高PI-IBS大鼠的内脏痛觉高敏感性压力阈值,减少大鼠排稀便次数(P<0.05,P<0.01);防风可调节肠道菌群,显著降低粪便丝氨酸蛋白酶活性,抑制大鼠结肠黏膜PAR-2 mRNA基因表达及增加紧密连接ZO-1蛋白表达。结论防风具有显著的止痛、止泻效应,调节菌群和抑制肠道丝氨酸蛋白酶信号是其可能的作用机制。
Objective To study the effect of Radix Saposhnikoviae(RS) on intestinal flora and serine protease signal in rats with post-infection irritable bowel syndrome(PI-IBS). Methods PI-IBS rat model was established by intragastric administration of campylobacter. And then we observed the analgesic and antidiarrheal effect of RS and the effect of RS on intestinal flora, dung serine protease activity, colon protease activation receptor 2(PAR-2) mRNA expression and tight junction ZO-1 protein expression. Results RS increased the intestinal pressure threshold, and reduced the defecation frequency of PI-IBS rats(P 〈 0.05, P 〈 0.01 ). RS regulated the intestinal flora, significantly decreased the serine protease activity, inhibited the PAR-2 mRNA expression in intestinal mucosa, and increased the tight junction ZO-1 protein expression. Conclusion RS has significant analgesic and antidiarrheal effect. Regulation of intestinal flora and inhibition of the serine protease signal probably contribute to the therapeutic mechanism of RS.
