银杏黄酮苷元对小鼠脑缺血模型和鹌鹑动脉粥样硬化模型的影响 被引量:5

Effects of Ginkgetin Aglycone on Mouse Cerebral Ischemia Model and Quail Atherosclerosis Model

摘要 目的研究银杏黄酮苷元(GFA)抗脑缺血、动脉粥样硬化形成及降血脂的药理学活性。方法通过夹闭小鼠双侧颈总动脉复制脑缺血模型和高脂饲料饲喂鹌鹑复制动脉粥样硬化模型,检测GFA对小鼠喘息时间、脑组织含水量、丙二醛(MDA)、超氧化物歧化酶(SOD)和一氧化氮(NO)含量变化以及鹌鹑动脉粥样硬化进程中脂斑块形成和血脂[总胆固醇(TC)、甘油三脂(TG)、高密度脂蛋白(HDL)和低密度脂蛋白(LDL)]水平变化的影响。结果 GFA可延长小鼠夹闭双侧颈总动脉后喘息时间,降低脑含水量,减少MDA和NO的生成,提高SOD的活力(P<0.05,P<0.01);并且200 mg·kg-1 GFA预防组和中剂量组可降低动脉粥样硬化脂斑块发生率(P<0.05,P<0.01),100 mg·kg-1 GFA组TC、LDL含量降低(P<0.05)。结论 GFA对脑缺血和动脉粥样硬化具有较好的防治作用,为GFA防治心脑血管疾病提供了实验数据。 Objective To study the protective effects of ginkgetin aglycone (GA) on the development of cerebral ischemia in mice and atherosclerosis (AS) in quails, and to investigate the pharmacological action of decreasing blood lipids. Methods Mouse cerebral ischemia model was established by clamping of bilateral common carotid arteries, and quail AS model was set up by feeding high fat forage. And then we observed the effects of GA on gasp time, brain water content, and serum contents of malondialdehyde (MDA), super oxide dismutase (SOD) and nitric oxide (NO). The development of plaque and serum levels of total cholesterol (TC), triglycerides(TG), high-density l ipoprotein cholesterol (HDL-C) and low-density lipoprotein cholesterol (LDL-C) in quail model were also monitored. Results Compared with the model group, GA could prolong the time of gasp, reduce MDA, NO and brain water content, and increase the activity of SOD (P 〈0.05 or P 〈0.01). Furthermore,GA pretreatment group and middle-dose GA treatment group in the dosage of 200 mg.kg-1 could significantly delay the development of plaque (P 〈 0.05 or P 〈 0.01), and 100 mg.kg-1 GA treatment group reduced TC and LDL-C levels(P 〈 0.05). Conclusion GA shows certain therapeutic and preventive effect on cerebral ischemia and AS, which will supply evidence for the development of GA into a drug for cardia-cerebrovascular disease.
