AQP4及AQP4 mRNA在重型脑损伤中脑脊液中的表达及意义

摘要 目的观察重型脑损伤后脑脊液中水通道蛋白(4AQP4)及AQP4mRNA的表达变化及意义。方法选取重型颅脑损伤患者51例,对每例患者第0、3、7天脑脊液进行AQP4ELIS(A酶联反应吸附实验)含量测定,RT-PC(R实时定量聚合酶链式反应)测定AQP4mRNA表达。结果第0、3、7天脑脊液中均可检测AQP4及AQP4mRNA。第3天较第0天升高(P<0.05),第7天较第3天降低(P<0.05),第7天与第0天比较,差异无统计学意义。AQP4与AQP4mRNA相关(r=0.867)。结论重型脑损伤后患者脑脊液AQP4的表达随着病程变化,推测AQP4可能参与了脑损伤的病理过程,与病情的转归相关。 Objective To observe the change of aquaporin (AQP)4 and AQP4 mRNA level in the cerebrospinal fluid of patients with severe brain injury and its clinical significance.Methods There are 51 severe traumatic brain injury patients. ELISA (enzyme-linked immunosorbent assay) was used to determine cerebrospinalfluid AQP4 protein expression after 0,3,7 days,and RT-PCR (reverse transcription PCR) was used to determine AQP4 mRNA expression.Results AQP4 protein or mRNA expressed within the 0,3,7 days.The level of AQP4 or AQP4 mRNA increased 3 days after operation (P <0.05),and then statistically decreased 7 days after operation (P <0.05). But the 7 days had no statistically difference compared with 0 day after operation.Conclusion After Brain injury, AQP4 expression changes after brain injury is closely relatedwith the formation of cerebral edema, AQP4 in the damage brain edema formation plays an important role.
