摘要
目的探讨十溴联苯醚(decabromodiphenylether,BDE-209)与邻苯二甲酸二(2-乙基)酯(Bis(2-ethylhexyl)phthalate,DEHP)联合暴露致青春期雄性小鼠生殖损伤的表现。方法 4周龄青春期雄性ICR小鼠随机分为5组:A组为玉米油对照组;B组为100 mg/(kg·d)BDE-209+100 mg/(kg·d)DEHP组;C组为100 mg/(kg·d)BDE-209+300 mg/(kg·d)DEHP组;D组为300 mg/(kg·d)BDE-209+100 mg/(kg·d)DEHP组;E组为300mg/(kg·d)BDE-209+300 mg/(kg·d)DEHP组。小鼠经口染毒6周后处死,称取小鼠脏器重量并计算脏器系数,附睾尾部精子计数,采用放射免疫检测法检测小鼠血清睾酮浓度,采用免疫印迹实验检测小鼠睾丸睾酮合成酶的表达水平。结果与对照组相比,E组小鼠睾丸脏器系数降低(P<0.001),D、E组小鼠血清睾酮浓度显著降低(P=0.021,P=0.003),各联合暴露组小鼠附睾精子计数均降低(均有P<0.001),且小鼠睾丸类固醇激素合成急性调节蛋白St AR、胆固醇侧链裂解酶(P450scc)、17β-羟基类固醇脱氢酶(17β-HSD)合成表达量降低。结论 BDE-209和DEHP联合暴露,能够诱导青春期雄性ICR小鼠雄性生殖功能损伤。
Objective To discuss the reproductive toxicity of BDE-209 and DEHP combined exposure to the adolescent male ICR mice. Methods The 4 weeks old adolescent male ICR mice were randomly divided into 5 groups: A: corn oil control group, B:100 mg/(kg · d) BDE-209 + 100 mg/(kg·d) DEHP group; C:100 mg/(kg·d) BDE-209 + 300 mg/(kg·d) DEHP group; D:300 mg/(kg·d) BDE-209 +100 mg/(kg·d) DEYIP group;and E:300 mg,/(kg·d) BDE-209 +300 mg,/(kg·d) DEHP group. The mice were executed 6 weeks after BDE-209 and DEHP combined exposure. The mice sperm' s number was recorded and the testosterone tested by radioimmunoassay. The levels of STAR, P450scc, and 1713-HSD expression in testicular were measured by Western blot. Results Compared with the control group, the E group mice testicular coefficient was significantly decreased ( P 〈 0. 001 ). The D and E group mice Lg( serum testosterone, 2. 14 ± 0. 84 ng/mL, 2. 06 ±0. 67 ng/mL respectively ) was decreased (P = 0. 021,P = 0. 003 ) and all the combined exposure groups mice sperm's number( × 10^6/mL) was decreased (P 〈0. 001 ). The STAR, P450scc and 17β- HSD expressions in testicular were down-regulated. Conclusions BDE-209 and DEHP combined exposure could induce reproductive impairment in the adolescent male ICR mice.
