摘要
以显微注射的方法,将去掉原核DNA的Pc DNA3.1IGFTREtr TA诱导型IGF-1载体注射到猪受精卵原核中,经胚胎移植、怀孕、产仔、取样、检测等过程,得到幼仔85头,其中的11头为PCR检测阳性,进一步的Southern blot检测有10头阳性,即构建的原代诱导型IGF-1转基因猪有10头。
In this study, the pc DNA3.1IGFTREtr TA induced IGF-1 vector without prokaryotic DNA was microinjected into pronucleus of pig fertilized eggs. Through embryo transfer, pregnancy, parturition, sampling and testing procedures, 85 little life born, of which 11 was tested positive for PCR, 10 positive for Southern blot detection. It indicated that there had 10 primary transgenic pigs of inducible IGF-1 in the test.
