电针对功能性消化不良肝郁脾虚型大鼠中枢及外周VIP及其受体VPAC-1的影响 被引量:19

Effects of electro-acupuncture on central and peripheral VIP and its receptor VPAC-1 expression in functional dyspepsia rats with syndrome of liver stagnation and spleen deficiency

摘要 目的:观察电针功能性消化不良(FD)肝郁脾虚模型大鼠中枢及外周血管活性肠肽(VIP)及其受体(VPAC1)的影响。方法:48只SD大鼠随机分为空白组、模型组和电针组。除空白组外均采用多因素干预法造模,造模成功后对电针组大鼠进行电针治疗28d,结束后对3组大鼠进行灌胃及解剖取材,测定胃内残留率及小肠推进率;Western blot法测定大鼠下丘脑、胃窦、空肠的VIP、VPAC1表达。结果:与空白组相比,模型组大鼠VIP、VPAC1的表达及胃内残留率均明显升高(P<0.05,P<0.01),小肠推进率显著降低(P<0.01);与模型组相比,电针组大鼠VIP、VPAC1的表达及胃内残留率均明显降低(P<0.05,P<0.01),小肠推进率显著升高(P<0.01)。结论:电针能够显著地降低外周,特别是中枢VIP、VPAC1的水平,有效改善胃肠动力障碍,提示电针是作用于脑肠轴调节脑肠肽的分泌,从而调节胃肠道活动,这可能是电针治疗FD的作用机制。 Objective: To observe the effects of electro-acupuncture(EA) on central and peripheral VIP and its receptor VPAC-1 expression in functional dyspepsia(FD) rats with syndrome of liver stagnation and spleen deficiency. Methods: Fortyeight rats were randomly divided into three groups: control group, model group and EA group. Except for the control group, the other groups suffered from multi-factor intervention model. Rats in the EA group were given the treatment of EA for consecutive 28 days. Then, all rats were sacrificed to detect gastric remnant rate and intestinal movement rate. The expression of VIP and VPAC-1 in hypothalamus, gastric antrum and empty intestine were detected with Western blot. Results: Compared with the control group, the expression of VIP, VPAC-1 and gastric remnant rate in model group were increased(P<0.05, P<0.01), but intestinal movement rate was significantly decreased(P<0.01). Compared with the model group, the expression of VIP, VPAC-1 and gastric remnant rate in EA group were decreased(P<0.05, P<0.01), but intestinal movement rate was significantly increased(P<0.01). Conclusion: The treatment of electro-acupuncture could significantly decrease the expression of VIP and VPAC-1 in periphery and central nervous system and improve gastrointestinal motility. It indicates that the roles of electro-acupuncture in the secretion of brain-gut peptide through the brain gut axis could regulate the gastrointestinal activity, which may be mechanism underlying electro-acupuncture treating functional dyspepsia.
