白僵蚕多糖的提取工艺条件优化及抗氧化活性测定 被引量:9

Extraction Process Optimization and Antioxidant Activity Determination of Polysaccharide from Bombyx Batryticatus

摘要 多糖是白僵蚕的重要活性成分之一。采用水提醇沉法提取白僵蚕的多糖组分,在单因素试验的基础上,选择提取温度、提取时间和原料质量浓度3个因素设计Box-Benhnken中心组合试验,以白僵蚕多糖得率为响应值进行响应面分析优化提取工艺条件,并考察白僵蚕多糖提取物的体外抗氧化活性。优化的白僵蚕多糖提取工艺条件为提取温度100℃、提取时间160 min、原料质量浓度40 g/L,在此条件下白僵蚕多糖的得率为4.43%,提取液经醇沉干燥后所得粗多糖纯度为49.22%。获得的白僵蚕多糖提取物具有较好的抗氧化活性,且总还原力和清除羟自由基能力均呈现一定的量效关系。建立的白僵蚕多糖提取工艺操作简单、提取效果较好,白僵蚕多糖的抗氧化活性有利于白僵蚕在中成药或功能食品方面的深度开发利用。 Polysaccharide is an important functional compound of Bombyx batryticatus. In present study, polysaccha- ride composition in Bombyx batryticatus was extracted by using hot water extraction and alcohol precipitation method. Based on single-factor experiments, three factors including extraction temperature, extraction time and mass concen- tration of raw material were selected to conduct Box-Benhnken central composite design. Taking yield of polysacchari- de as response value, the extraction process conditions were optimized through response surface analysis. Besides, in vitro antioxidant activity of polysaccharide extract from Bombyx batryticatus was determined. The optimal process con- ditions for extracting polysaccharide from Bombyx batryticatus were extraction temperature 100℃, extraction time 160 min and mass concentration of raw material 40 g/L. Under this optimized condition, the yield of polysaccharide was 4.43%, and the purity of crude polysaccharide obtained from extraction solution by alcohol precipitation method was 49.22%. The obtained polysaccharide extract from Bombyx batryticatus exhibited high antioxidant activity, and the total reducing capacity and hydroxyl radical scavenging activity were concentration dependent. The optimized extraction process is easy to operate and favorable to the extraction of polysaccharide from Bombyx batryticat- us. In addition, the antioxidant activity of polysaccharide would facilitate the deep development and utilization of Bombyx batryticatus in the fields of Chinese patent medicine and functional food.
