梅花鹿瘤胃液中棒状杆菌的分离鉴定与系统发育树的构建 被引量:2

摘要 营养琼脂平板上需氧、厌氧培养梅花鹿瘤胃液中的细菌,挑取菌落,纯化培养,观察个体和菌落形态特征,同时进行生理生化特征鉴定;对细菌16Sr DNA测序,建立系统发育树。结果表明,分离出18株棒状杆菌科棒状杆菌属菌株,其中11株与瘤胃棒状杆菌同源性达99.93%以上,2株与嗜甘氨酸棒状杆菌同源性达99.71%,5株与极小棒状杆菌同源性为96.83%;结合生理生化鉴定结果,分离的18株细菌均为棒状杆菌科棒状杆菌属细菌。 16Sr DNA sequencing and systematic phylogenetic tree were processed from the single clony cloning of bacterias of sika deer rumen fluid,which were cultured in agar plate by aerobically and anaerobically,and characterized physiologically and biochemically. The results showed that 18 strains of bacterias were identified as members of Corynebacterium bacillus; 11 out of 18 were homologous to C.vitaeruminis by 99. 93%; and 2 out of 18 were homologous to C. glycinophilum by 99. 71%; 5 out of 18 were homologous to C. minutissimum by 96. 83%.
