摘要
【目的】探讨越橘茎段离体培养及试管苗出瓶移栽技术,为广东地区越橘试管苗规模化生产提供技术支持。【方法】以高丛越橘品种夏普蓝和兔眼越橘品种梯芙蓝带芽茎段为外植体进行离体培养,筛选适合腋芽诱导、丛生芽增殖、植株再生和生根的培养基;在试管苗出瓶移栽阶段进行栽培基质、光照和温度条件的优化筛选。【结果】两个越橘品种茎段在初代培养基WPM+ZT0.5mg/L中的萌芽率分别为77.8%和63.9%,分别比在MS+ZT0.5mg/L中高27.8%和16.7%;夏普蓝在增殖培养基WPM+ZT1.0mg/L中不定芽增殖达11.7倍,梯芙蓝在增殖培养基WPM+ZT2.0mg/L中增殖最高达10.3倍;夏普蓝在生根培养基WPM+IBA0.5mg/L中生根率达91.7%,梯芙蓝在生根培养基WPM+IBA1.0mg/L中生根率达88.9%。夏普蓝和梯芙蓝组培苗在泥炭土+河沙(2:1)基质中的移栽成活率分别为91.7%和88.3%;在透光20%-40%及25℃条件下移栽,成活率、株高和茎粗等指标均达最优水平。【结论】广东地区已具备越橘夏普蓝和梯芙蓝组培苗规模化生产条件。
[Objective ]The stem explants in vitro culture and plantlet transplanting technique of Vaccinium were in- vestigated, in order to provide technical supports for its large-scale rapid propagation in Guangdong region. [ Method ]Us- ing stems of Vaccinium corymbosum Sharpblue and V. asbei Tifblue as explants, the optimal media for bud induction, tufty bud multiplication, plantlet regeneration and rooting were screened. Then the culture substrates, illumination and temperature conditions were optimized during plantlet transplanting period. [Result]The results showed that, the germina- tion rates of stem explants of two Vaccinium varieties in WPM medium with 0.5 mg/L ZT were 77.8% and 63.9%, respec- tively, which were 27.8% and 16.7% higher than those in MS medium with 0.5 mg/L ZT medium. And the multiplication times of V. corymbosum Sharpblue adventitious buds reached up to 11.7 in WPM multiplication medium with 1.0 mg/L ZT. While the multiplication time of V. ashei Tifblue reached up to 10.3 in WPM medium with 2.0 mg/L ZT. The rooting rate of V. corymbosum Sharpblue reached up to 91.7% in WPM rooting medium with 0.5 mg/L IBA, while that of V. ashei Tifblue reached up to 88.9% in WPM rooting medium with 1.0 mg/L IBA. The transplantation survival rate of in vitro cultured plantlets of two Vaecinium varieties reached up to 91.7% and 88.3% in cultivation substrate made up of peat soil and sand (2:1), respectively. The survival rate, plantlet height, stem diameter and other indexes were optimal under 20%-40% light transmittance and ambient temperature of 25 ℃. [ Conclusion ]Guangdong region have sufficient conditions for large- scale production of V. corymbosum Sharpblue and V. ashei Tifblue test-tube plantlets.
