摘要
目的针对两种实验室常用全血基因组提取DNA试剂盒(天根血液基因组DNA提取试剂盒、康为世纪血液基因组提取试剂盒)提取方法和DNA产量进行比较,建立方便、快捷、有效的外周血液基因组DNA的提取方法。方法采用500ul、800ul全血、裂解液1.5倍、2倍提取基因组DNA,利用分光光度计和凝胶电泳检测提取效果。结果两种试剂盒采用800ul全血与2倍裂解液时提取血液基因组DNA浓度最高,其中天根试剂盒提取的基因组DNA纯度和质量更优。结论利用商品化试剂盒提取血液基因组DNA时,针对实验目的,可采取增加全血用量和提高裂解液体积的方法提高基因组DNA的产量和纯度。
Objective To establish a convenient, fast and efficient peripheral blood genomic DNA extraction method therefrom two kinds of whole blood genomic DNA extraction kitst (TIANGEN BIOTECH and KANGWEI BIOTECH), Methods Using 500 ul and 800ul whole blood with EDTA, and cracking liquid1.5 and 2 times volume to isolate ganomic DNA. UV spectrophotometer and gel electrophoresis were used to examine the results. Results 800 ul whole blood with 2 times cracking buffer are the most optimized conditions for two kinds of genomic DNA isolation kits. Conclusion especially the TIANGEN kit can extract more DNA with pure quanlity.
