摘要
从养殖柞蚕周边的土壤分离筛选和鉴定得到一株能够产生蛋白水解酶的菌株G1。通过单因素实验优化该菌株的培养基碳源、氮源、初始pH、发酵温度、发酵时间和接种量等产酶条件。实验结果表明,该菌株为Luteibacter anthropi。当培养温度为47℃,将菌株按4%接种量接种到以葡萄糖为碳源、蛋白胨为氮源、初始pH为8.0的培养基中发酵96h,在此最优发酵条件下产酶量达到1 520U/mL。
A strain G1 producing proteolytic enzyme was isolated and identified from surrounding soil of breeding tussah.Fermentation conditions for enzyme production including carbon source,nitrogen source,initial pH,temperature,time and bacterial load were optimized.Phylogenetic analysis and phenotypic characterization suggested strain G1 should be Luteibacter anthropi.The maximum enzyme activity of 1 520 U/mL was obtained when strain G1 was inoculated at 4% inoculum and incubated at 47℃and pH 8.0for 96 hwith glucose as carbon source and peptone as nitrogen source.
