螺旋藻多糖对糖尿病大鼠血糖及抗氧化作用的影响 被引量:10

Effect of Spirulina polysaccharide on blood glucose and antioxidant activity in diabetic rats

摘要 目的观察螺旋藻多糖对糖尿病大鼠的降糖作用,并从氧化性损伤角度探讨其作用机制。方法雄性SD大鼠腹腔注射链尿佐菌素制备糖尿病模型,将造模成功大鼠按照血糖值和体重随机分为糖尿病模型组、阳性对照组(格列本脲,20 mg/kg)、螺旋藻多糖高剂量组(200 mg/kg)和低剂量组(100 mg/kg),经口灌胃给药,另设正常对照组,时间为8周,观察螺旋藻多糖对糖尿病大鼠体重、血糖、超氧化物歧化酶(SOD)活力和丙二醛(MDA)含量的影响。结果模型组大鼠体重和血糖分别为(219.0±18.6)g,(27.6±2.4)mmol/L,血清SOD活力和MDA含量分别为(130.88±17.82)U/ml)、(11.24±0.93)nmol/ml;螺旋藻多糖高、低剂量组的体重和血糖分别为(273.0±26.9)g、(9.2±4.4)mmol/L和(270.1±43.4)g、(15.4±3.7)mmol/L,与模型组相比各剂量组大鼠体重明显增加(P<0.01),血糖显著下降(P<0.01),螺旋藻多糖高、低剂量组的SOD活力和MDA含量分别为(189.52±11.36)U/ml(5.87±0.61)nmol/ml和(155.72±19.0)U/ml、(6.65±1.02)nmol/ml,与模型组相比各剂量组血清中SOD活力均有提高(P<0.05),MDA含量显著下降(P<0.01)。结论螺旋藻多糖具有降血糖的作用,从氧化性损伤的角度分析,可认为其降糖的作用机制是通过增强SOD活力和降低MDA含量实现的。 [Objective]To observe the hypoglycemic effect of Spirulina polysaccharide on diabetic rats,explore the mechanism from the perspective of oxidative damage.[Methods]Male SD rats were treated with streptozotocin by intraperitoneal injection to get diabetes model.According to the blood glucose level and body weight,the diabetic rats were randomly divided into diabetic model group,positive control group(glyburide,20 mg/kg),high-dose Spirulina polysaccharide group(200 mg/kg) and low-dose Spirulina polysaccharide group(100 mg/kg),with the intragastrical administration for 8 weeks,and there was also a normal control group.The effects of Spirulina polysaccharide on body weight,blood glucose,SOD activity and MDA content in diabetic rats were observed.[Results]The body weight,blood glucose level,SOD activity and MDA content of diabetic model group was(219.0±18.6) gram,(27.6 ±2.4) mmol/L,(130.88 ±17.82) U/ml and(11.24±0.93) nmol/ml respectively.The body weight of high-dose Spirulina polysaccharide group and low-dose Spirulina polysaccharide group was(273.0±26.9) gram and(270.1 ±43.4)gram respectively,while the blood glucose level of two groups was(9.2±4.4) mmol/L and(15.4±3.7) mmol/L respectively.Compared with the model group,body weight in two Spirulina polysaccharide groups significantly increased(P〈0.01),and blood glucose level decreased significantly(P〈0.01).SOD activity of high-dose Spirulina polysaccharide group and low-dose Spirulina polysaccharide group was(189.52 ±11.36) U/ml and(155.72 ±19.0) respectively,while MDA content of two groups was(5.87±0.61) nmol/ml and(6.65±1.02) nmol/ml respectively.Compared with the model group,SOD activities in two Spirulina polysaccharide groups significantly increased,and MDA contents decreased significantly(P 0.01).[Conclusion]Spirulina polysaccharide has hypoglycemic effect.From the perspective of oxidative damage,it is considered that the mechanism is probably due to the increase of SOD activity and the decrease of MDA content.
