摘要
目的探讨染料木素对人肝癌细胞株HepG2增殖和凋亡的作用。方法体外培养HepG2细胞,分为对照组(A组)和不同浓度染料木素10、20、40、80、160μmol/L组(B1、B2、B3、B4、B5组)。采用细胞计数试剂盒(CCK-8)和流式细胞术分别检测HepG2细胞生长和凋亡,Hoechst33258染色法观察细胞形态学变化,Western blot法检测活化半胱天冬氨酸蛋白酶3(Caspase-3)的表达。结果与A组相比,染料木素呈浓度和时间依赖性地抑制细胞增殖,促进细胞凋亡,上调活化Caspase-3蛋白表达(P<0.01)。结论染料木素可能是通过诱导HepG2细胞凋亡抑制细胞生长。
Objective To investigate the effect of genistein on the proliferation and apoptosis of human hepatic cancer cell line HepG2. Methods HepG2 cells were cultured in vitro and divided into six groups of A(normal control), B1 (treated with genistein 10/amol/L), ]32 (treated with genistein 20 μmol/L), ]33 (treated with genistein 40 μmol/L), B4 (treated with genistein 80 μmol/L) and B5 (treated with genistein 160μmol/L). The cell viability and apoptosis were measured by CCK-8 assay and flow cytometry,respectively. The changes of cell morphology were observed by Hoechst 33258 staining. The protein expression of actived-Caspase-3 was detected by Western blot. Results Compared with group A,genistein could inhibit cell proliferation in the dose- and time-dependent manners, promote cell apoptosis and up-regulate the protein expression of actived Caspase-3 (P〈0. 01 ). Conclusion Genistein inhibits the proliferation of HepG2 cells probably via inducing cell apoptosis.
