摘要
[目的]研究冻干、冷冻、辐照对骨诱导活性的影响。[方法]收取胫骨上段松质骨,加工成大小约0.5 cm×0.5 cm×0.5 cm的骨块,随机分为五组:新鲜骨组、冷冻组、冻干组、冻干+辐照组以及冷冻+辐照组,用免疫组化法测定骨细胞因子BMP、bFGF、β-TGF含量的变化。[结果]新鲜骨及冷冻骨、冻干骨均有细胞因子表达,但冻干骨表达的程度着色较深,表达阳性的细胞位于骨小梁的表面。辐照后,BMP、bFGF、β-TGF表达减少。[结论]辐照可减少骨的诱导活性,但仍保留部分骨诱导成分。
[Objective]To investigate the effect of deep-frozen,freeze- dry and radiation on the ostoinduction properties of cancellous bone.[Method]The cancellous bone was cut from the proximal epiphy seal region and made into bone fragments with size of 5 × 5 × 5 mm^3.The all bone were divided into five groups randomly,ie fresh bone group,deep- frozen bone group,freeze- dry bone group freeze- dry bone plus radiation group and deep- frozen bone plus radiation group.The concent of the bone morphogenic protein(BMP),transform growth factor- β(TGF-β and basic fibroblast growth factor(BFGF) were measured with immunohistochemical staining(SABC method).[Result]The expression of BMP,β-TGF and BFGF were observed in the fresh bone and deep- frozen bone and freeze- dry bone.Expression of the cellular factors in freeze- dry bone were larger than those of the fresh bone and deep- frozen bone.The intensity of the positive stain of freeze- dry bone was more than those of the fresh bone and deep- frozen bone.The positive expressed cell was satiated in the surface of the bone trabeculac.The express of BMP,β-TGF and BFGF decreased after the bone had been irradiated.[Conclusion]The results suggest that iradiation can decrease the osteoinduction properties of bone.But the osteoinduction properties of bone will be still partly kept.
