摘要
一氧化氮(NO)是人体内一种关键的生物信号分子,它在神经传导、血压调节、免疫激活中扮演重要角色。神经型一氧化氮合酶(n NOS)产生的NO在正常生理浓度下有神经保护作用,但在病理状态下,n NOS过量产生的NO与多种神经退行性疾病、神经痛有密切的联系。选择性抑制n NOS有望治疗多种神经系统疾病。本文将介绍一些n NOS选择型抑制剂,重点介绍2-氨基吡啶类和噻吩-2-甲脒类n NOS抑制剂。
Nitric oxide( NO) plays a critical role in human body,and affects neuronal signaling,blood pressure and immune activation. NO generated by neuronal nitric oxide synthase( n NOS) is neuroproctective at normal physiological concentrations. However,in pathological conditions,NO associates with many neurodegenerative disorders as well as neuropathic pain. n NOS-selective inhibition is a promising therapeutic method for these disorders. This article will introduce some competitive n NOS-selective inhibitors,especially the compounds derived from 2-aminopyridines and thiophene-2-carboximidamides.
