摘要 目的:研究促性腺激素释放激素激动剂(GnRH-a)在体外作用于子宫内膜异位症患者的在位子宫内膜细胞,对其子宫内膜容受性相关指标HOXA10的表达影响。方法:使用亮丙瑞林(GnRH-a)、17β雌二醇+黄体酮,分别作用于子宫内膜异位症患者在位子宫内膜细胞及对照组正常子宫内膜细胞。观察其对子宫内膜间质细胞及腺上皮细胞HOXA10表达的影响。结果:经亮丙瑞林预处理后的E2+P实验组与单用E2+P的实验组比较,间质细胞HOXA10表达量升高,腺上皮细胞表达降低。对照组内比较,间质细胞及腺上皮细胞HOXA10表达量增加。结论:体外GnRH-a预处理对内异症在位子宫内膜细胞的局部作用,可以增加间质细胞HOXA10的表达,可能对黄体中期子宫内膜的胚胎容受性有一定改善。 Objective: To investigate the in vitro effects of gonadotropin-releasing hormone agonist (GnRH-a) on the expression of endometrial receptivity-related index HOXA10 through activation of eutopic endometrial cells of patients with en- dometriosis. Methods: Leuprorelin (GnRH-a) and 1713 estradiol+progesterone were used to treat eutopic endometrial ceils of patients with endometriosis and normal endometrial cells of control group respectively. Its effects on HOXA10 expression in endometrial stromal cells and glandular epithelial cells were observed. Results: Compared with experimental group that was treated with E2 +P alone, HOXA10 expression level in endometrial stromal cells of experimental group treated with E2 +P after leuprorelin pretreatment were signifiantly increased but expression in glandular epithelial cells were decreased. For comparison within control group, HOXA10 expression levels in endometrial stromal cells and glandular epithelial cells increased. Conclusion: Local effects of GnRH-a pretreatment in vitro on eutopic endometrial cells of endometriosis can increase expression of HOXA10 in stromal cells and might improve endometrial embryonic receptivity in mid-luteal phase to some degree.
