摘要
目的探讨骨桥蛋白(osteopontin,OPN)在人舌鳞状细胞癌Tca-8113细胞上皮-间质转化(epithelial-mesenchymal transition,EMT)现象中的调控作用及其对Tca-8113细胞生物学行为的影响。方法 Tca-8113细胞稳定转染OPN,RT-PCR和Western blot技术检测转染效率;Western blot及免疫细胞化学法检测转染前后OPN、Snail及EMT标志物的变化;通过细胞黏附、迁移及Transwell侵袭实验,观察OPN对Tca-8113细胞黏附、迁移和侵袭等生物学行为的影响。结果 Tca-8113细胞能够稳定表达OPN;与转染前相比,转染后上皮标志物E-cadherin、β-catenin表达明显降低,而间质标志物vimentin、N-cadherin及Snail表达增加;Tca-8113细胞黏附、迁移和侵袭能力明显增加(P<0.05)。结论 OPN增强人舌鳞状细胞癌Tca-8113细胞黏附、迁移和侵袭能力,可能与其促进口腔鳞状细胞癌EMT的发生有关,Snail可能介导OPN的作用。
Purpose To investigate the effect of OPN on EMT phenomenon and the biological behavior of Tca-8113 cells lines. Methods Tca-8113 cell was stably transfected with a plasmid OPN,in order to establish OPN overexpression Tca-8113 cell lines,and the transfect efficiency was detected by Western blot and RT-PCR assays. Western blot and cell immunohistochemistry were used to detect OPN,Snail and EMT markers. Cell adhesion assay,migration assay and Transwell invasion assay were used to observe the effect of OPN on cell adhesion,migration and invasion of Tca-8113. Results Tca-8113 cells can stably expressed OPN. Expression of epithelial markers( E-cadherin,β-catenin) compared with the before transfection OPN were decreased,but mesenchymal markers( vimentin,N-cadherin) were increased. Snail expression also increased after transfection. After transfection of OPN,cell adhesion,migration and invasion of Tca-8113 were increased( P 〈 0. 05). Conclusion OPN is involved in promoting Tca-8113 cells adhesion,migration and invasion by regulating EMT phenomenon,in which Snail may be involved in this process.
