外周血单个核细胞VEGF基因在糖尿病患者中的表达及其与糖尿病肾病进展的相关性 被引量:2

The expression of VEGF gene in the diabetes patients and its correlation with the kidney disease progress caused by the diabetes

摘要 目的探讨糖尿病患者外周血单个核细胞(PBMCs)中血管内皮生长因子(VEGF)基因表达变化,分析其与糖尿病肾病进展的相关性。方法选取本院内分泌科2013年-2014年51例糖尿病患者作为实验组,50例健康体检者作为对照组,real-time PCR法检测糖尿病患者PBMCs VEGF基因mRNA表达水平,酶联免疫吸附试验(ELISA)测定血清中VEGF水平。结果与对照组比较,糖尿病组VEGF基因mRNA表达升高,血清VEGF水平也升高,差异有统计学意义(P<0.05)。糖尿病PBMCs VEGF基因mRNA表达量随着糖尿病肾病严重程度依次升高:DN3>DN2>DN1>DN0,而DN3组、DN2组血浆VEGF水平显著高于DN1组、DN0组,差异有统计学意义(P<0.05)。结论糖尿病患者PBMCs VEGF基因表达显著升高,与糖尿病肾病发生发展呈正相关,或可成为监测糖尿病肾病进展又一靶标。 Objective To investigate the vascular endothelial growth factor( VEGF) expression changes of peripheral blood mononuclear cells( PBMCs) from the diabetes patients and the relationship with the process of kidney disease caused by the diabetes. Methods 51 diabetes patients were selected from the endocrinology department in our hospital as the test group during2013- 2014. Moreover,we select 50 healthy subjects as the control group. The VEGF mRNA expression in PBMCs in diabetes patients was detected by real time- PCR. The serum VEGF levels were detected by the ELISA method. Results Compared with the control group,the mRNA expression level of the VEGF gene in the diabetes group significantly reduced,so did the serum VEGF level,and the difference was statistically significant( P〈0. 05). The mRNA expression of VEGF gene in PBMCs increased by turn: DN3 〉DN2 〉DN1 〉DN0. However,the plasma VEGF levels in the DN3 and DN2 groups were significantly higher than the DN1,DN0 groups,and the differences were statistically significant( P〈0. 05). Conclusion The levels of the VEGF expression in peripheral white blood cell increased in the diabetes patients. In addition,there was a positive correlation with the progress of the kidney disease caused by the diabetes. It may be one of the main markers to monitor the progress of the kidney disease caused by the diabetes.
