维生素D受体基因BsmⅠ多态性与新疆维吾尔族、汉族儿童急性白血病的相关性研究 被引量:1

摘要 目的探讨维生素D受体(VDR)基因BsmⅠ位点单核苷酸多态性与新疆维吾尔族、汉族儿童急性白血病(AL)的相关性。方法以新疆医科大学第一附属医院儿科150例急性白血病患者(维吾尔族83例,汉族67例)及161例(维吾尔族75例,汉族86例)同期健康体检儿童为研究对象,用聚合酶链反应-限制性片段长度多态性分析(PCR-RFLP)检测VDR基因BsmⅠ多态性分布。结果 1白血病患儿与对照组的BsmⅠ基因型、等位基因分布比较,差异有统计学意义(χ2=9.326和5.983,P=0.009和0.014);2对于维吾尔族儿童,病例组与对照组的BsmⅠ基因型(GG+GA/AA)、等位基因分布比较,差异无统计学意义(χ2=1.296和1.027,P=0.255和0.311);而对于汉族儿童,病例组与对照组的BsmⅠ基因型(GG+GA/AA)、等位基因分布比较,差异有统计学意义(χ2=10.414和6.661,P=0.001和0.010)。3就白血病患儿而言,维吾尔族与汉族的BsmⅠ基因型、等位基因分布比较,差异无统计学意义(χ2=7.797和2.518,P=0.180和0.113);对照组中维吾尔族与汉族的BsmⅠ基因型、等位基因分布差比较,差异无统计学意义(χ2=0.650和0.000,P=0.420和0.993)。结论白血病患儿与非白血病患儿VDR基因BsmⅠ的多态性分布区别不大,BsmⅠ的多态性分布与白血病有相关性。其中BsmⅠ基因分布与汉族儿童白血病有相关性,而与维吾尔族儿童白血病无关。无论在白血病患儿或正常儿童,BsmⅠ基因型分布都不存在民族差异。 [Objective] To investigate the relationship between vitamin D receptor (VDR) gene Bsm I polymorphism and hereditary susceptibility of Han and Uighur children with acute leukemia. [Methods] The VDR gene Bsm I was detected in 150 children with acute leukemia (83 Uighur and 67 Han) and 161 healthy controls (75 Uighur and 86 Han) using polymerase chain reaction-restriction fragment length polymor- phism method. [Results ] ①There were statistically significant differences in frequencies of Bsm I genotype and the alleles between the acute leukemia cases and healthy controls (X2= 9.326 and 5.983, P= 0.009 and 0.014). ②There was no significant difference in frequencies of Bsm I genotype (GG+GA/AA) or the alleles between the acute leukemia cases and healthy controls of Uighur children (X2= 1.296, 1.027; P=0.255, 0.311), while there were statistically significant differences in frequencies of Bsm [ genotype (GG+GA/AA) and the alleles between the acute leukemia cases and healthy controls of Han children (X2= 10.414 and 6.661, P =0.001 and 0.010). ③There was no significant difference in frequencies of Bsm I genotype or the alleles of either acute leukemia cases (χ2= 7.797, 2.518; P= 0.180, 0.113) or healthy controls (χ2= 0.650, and 0.000; P= 0.420 and 0.993) between the Han and Uighur children. [Conclusions] Polymorphism distribution of VDR gene Bsm I is associated with acute leukemia of Han children, but not associated with acute leukemia of Uighur children. Bsm I is not associated with ethic difference.
