摘要
为从贯叶金丝桃中获得含量较高的金丝桃苷,并克服传统提取法效率低、溶剂消耗多、耗时长等问题,采用酶法提取贯叶金丝桃中金丝桃苷成分。在单因素试验基础上,选取纤维素酶用量、酶解时间、酶解温度以及酶解pH进行4因素3水平的响应面试验。结果表明:提取金丝桃苷的最佳工艺为乙醇浓度60%、液固比25∶1、纤维素酶用量5.7mg/g、酶解时间106min、酶解温度51℃、酶解pH4.6,在该条件下贯叶金丝桃中金丝桃苷的提取率达91.41%。
The enzymatic extraction method was tested to overcome the disadvantages including low efficiency,high solvent consumption and longer time-consuming of the traditional extraction method and improve hyperin extraction rate from Hypericum perforatum.The regression equation was established by the 4factors with three levels design based on single-factor test.Results:The hyperin extraction rate from Hypericum perforatum under the optimum extraction conditions of 60% ethyl alcohol,25∶1of liquid/solid ratio,5.7mg/g cellulase,pH 4.6and at 51℃for 106 min can reach 91.41%.
