摘要
目的探讨回药爱康方含药血清对Lewis肺癌细胞(LLC)增殖及细胞周期的影响。方法雄性SD大鼠80只,按体重随机分为8组:生理盐水对照组(NS),回药爱康方低、中、高剂量(AKL、AKM、AKH)组,环磷酰胺(CTX)组,回药爱康方低、中、高剂量联合环磷酰胺(AKL/AKM/AKH+CTX)组。各组同时相应给予回药爱康方低、中、高剂量浓缩水煎剂灌胃,连续5d;CTX组和联合组再按20mg·kg-1给予环磷酰胺于第1、3、5天腹腔注射,NS组给予等量生理盐水。制备各含药血清,体外培养LLC。应用MTT法检测各组含药血清对LLC增殖的影响,流式细胞仪进行细胞周期时相分析。结果回药爱康方含药血清能够抑制LLC增殖,各含药血清组与NS组比较差异有统计学意义(P<0.05)。其中高剂量联合CTX组的抑制率明显高于其他组,抑制程度呈时间和浓度依赖性;流式细胞仪检测结果显示,回药爱康方含药血清能使LLC细胞阻滞于G1期,随着药物浓度的增加,G1期细胞增多、S期细胞减少,高剂量以及联合组尤为明显(P<0.05)。结论回药爱康方含药血清可抑制LLC生长增殖,并使细胞阻滞于G1期,提示回药爱康方含药血清可能通过阻滞细胞周期,干扰DNA合成而抑制肺癌细胞的增殖。
Objective To investigate the effect of medicated serum contained with Hui medicine Ai Kang recipe on cell cycle of LLC. Methods 80 SD rats were randomly divided into eight groups according to the weight,normal saline( NS) group,low and medium and high dose Hui medicine Ai Kang recipe,cyclophosphamide( CTX),low and medium and high dose Hui medicine Ai Kang recipe combined with CTX. At the same time rats took Hui medicine Ai Kang recipe,cyclophosphamide and normal saline for 5 days. Effect of medicated serum contained with Hui medicine Ai Kang recipe on growth and proliferation of LLC were detected by MTT. The target cells were harvested to analyze the cell cycles by flow cytometry. Results High dose Hui medicine Ai Kang recipe could significantly inhibit the proliferation of LLC. The inhibition rate in High dose Hui medicine Ai Kang recipe group was significantly higher than that in other groups. Flow cytometry showed that LLC were blocked at G1 stage after drug treatment. In a dose dependent manner,G1 stage cells increased while S stage cells decreased with the concentration increase of Hui medicine Ai Kang recipe( P〈0. 05).Conclusion Hui medicine Ai Kang recipe can significantly inhibit the proliferation of LLC and LLC were blocked at G1 stage after drug treatment. The antineoplastic mechanism of Hui medicine Ai Kang recipe may be due to blocking cell cycle and inhibiting DNA synthesis to restrain LLC proliferation.
