参附注射液对阿霉素所致心衰大鼠氧化应激水平及血管紧张素Ⅱ的影响 被引量:18

摘要 目的探讨参附注射液对阿霉素(ADR)所致心衰大鼠氧化应激水平及血管紧张素Ⅱ(AngiotensinⅡ)的影响。方法将90只大鼠随机分为6组:正常对照组、模型组、卡托普利组(作为阳性对照),参附注射液低、中、高剂量组,每组各15只。正常对照组仅给予等体积生理盐水,模型组于实验开始后第2、4天腹腔注射ADR 1 mg/kg,第6、8天腹腔注射ADR 2 mg/kg,第10、12天腹腔注射ADR 3 mg/kg,第14、16天腹腔注射ADR 4 mg/kg,累计用药剂量达20 mg/kg。卡托普利组于实验开始后第5天卡托普利180 mg/(kg·d)灌胃。参附组于造模后1周,按低、中、高剂量组分别腹腔注射参附注射液1、2、4 mL/kg,每日1次,共2周。处理结束后测定心肌超氧化物歧化酶(SOD)活力和心肌丙二醛(MDA)含量;测定血清中AngiotensinⅡ水平。光镜观察心肌病理形态学变化;电镜观察心肌超微结构变化。结果阿霉素能显著降低大鼠心肌SOD活性,卡托普利、4 mL/kg参附注射液可将心肌中SOD活性升高至(125.26±7.21)、(128.44±10.02)U/L,与模型组比较差异有高度统计学意义(P<0.01);ADR也提高了大鼠心肌MDA含量和血清AngiotensinⅡ水平,而给予卡托普利、4 mL/kg参附注射液处理后,大鼠心肌中MDA含量分别降至(8.82±0.16)、(8.48±0.28)nmol/mg,血清AngiotensinⅡ水平降至(388.32±6.88)、(395.21±10.02)ng/L,与模型组比较,差异有高度统计学意义(P<0.01),而且心肌的病理学形态有了很大改善。结论参附注射液可明显改善心衰大鼠的心功能,保护机制可能与抗氧化及降低AngiotensinⅡ的含量有关。 Objective To explore the effects of Shenfu Injection on oxdative stress and angiotensin Ⅱ of rats with heart failure induced by Adriamycin.Methods Totally 90 rats were randomly divided into five groups with 15 rats in each group:normal control group,model control group,Captopril group(as positive control),and Shenfu Injection low,medium,high dose group.The normal control group was only treated with saline by intraperitoneal injection throughout the course of the experiment,while the other four groups were given Adriamycin by intraperitoneal injection with the total dosage of 20 mg/kg.For the Shenfu Injection low,medium,high dose group,1,2,4 mL/kg Shenfu Injection were given after Adriamycin Injection seven days later.For the positive control group,180 mg/(kg·d) Captopril was given.After the model was successfully established,relevant indexes were measured.The activities of plasma superoxide dismutase(SOD) and the contents of malondialdehyde(MDA) in myocardial tissues were measured.The levels of angiotensin Ⅱ in serum were detected.Pathological changes in cardiomyocyte were observed.With electron microscope,the cardiac ultrastructural changes in damaged myocardium were examined.Results Adriamycin significant decreased the activities of SOD.After treatment with Captopril and 4 ml/kg Shenfu Injection,the activities of superoxide dismutase increased to(125.26±7.21),(128.44±10.02) U/L.There were great significances as compared with those in model group,the differences were statistically significant(P0.01).Adriamycin also increased the contents of MDA and the levels of angiotensin Ⅱ.After treatment with Captopril and 4 mL/kg Shenfu Injection,the contents of MDA in myocardial tissues decreased to(8.82±0.16),(8.48±0.28) nmol/mg.The levels of angiotensin Ⅱ in serum decreased to(388.32±6.88),(395.21±10.02) ng/L,compared with those in model group,the differences were statistically significant(P〈0.01).In addition,the changes of myocardial pathological and ultrastructural damages were improved.Conclusion Shenfu Injection significantly improves the cardiac function of rats with adriamycin-induced heart failure through antioxidant and reducing the angiotensin Ⅱ level.
