摘要
目的探讨乳杆菌对小肠上皮钙离子吸收的影响及其作用机制。方法测定植物乳杆菌ST III发酵上清液对复合钙盐解离效果的基础上,通过MTT比色法测定植物乳杆菌ST III发酵上清液对大鼠小肠隐窝上皮细胞(IEC-6)增殖率影响以及用fluo-3AM荧光染色观察对IEC-6细胞内游离钙离子浓度变化。结果植物乳杆菌ST III发酵上清液对大鼠IEC-6增殖有明显的促进作用,与对照组比较差异有统计学意义,24h(F=548.401,P=0.000,P<0.05),48h(F=722.352,P=0.000,P<0.05),并呈良好的量效关系;同时植物乳酸菌ST III发酵上清液刺激后小肠隐窝上皮细胞6、12和24h,细胞内游离钙离子荧光强度相对值明显高于对照组,6h(F=203.911,P=0.000,P<0.05),12h(F=106.158,P=0.000,P<0.05),24h(F=26.076,P=0.000,P<0.05),差异有统计学意义。结论植物乳杆菌ST III在细胞水平上促进小肠上皮增殖并对细胞内Ca2+有明显的促进作用。
Objective To discuss the effect and mechanism of Lactobacillus plantarum ST IIIon calcium absorption.Methods Effects of Lactobacillus plantarum spent culture supernatant on proliferation rates of rat small intestinal crypt epithelial(IEC-6)cells were determined by MTT,based on the dissociation action of Lactobacillus plantarum on insoluble calsium salt.Cellular calcium concentrations in IEC-6cells which treated by Lactobacillus plantarum spent culture supernatant were observed by fluorescence microscopy(fluo-3AM).Results Lactobacillus plantarum spent culture supernatant could apparently promote IEC-6cell proliferation by dose-dependent way.Compared with the control group the difference was statistically significant,24h(F=548.401,P=0.000,P〈0.05),48h(F=722.352,P=0.000,P〈0.05).Intracellular IEC-6cells calcium concentration was increased after Lactobacillus plantarumspent culture supernatant stimulation.The results showed that the fluorescence intensity of free calcium in the small intestine was significantly higher than that of the control group after 6h(F=203.911,P=0.000,P〈0.05),12h(F=106.158,P=0.000,P〈0.05)and 24h(F=26.076,P=0.000,P〈0.05).Conclusion This strain of Lactobacillus plantarumt ST III have a significant effect on the absorption of Ca2+in the cell level.
