摘要
目的比较第6、7、8肋软骨以及耳软骨的Ⅱ型胶原蛋白含量。方法利用RT-PCR检测方式测得Ⅱ型胶原蛋白m RNA在第6、7、8肋软骨和耳软骨中的表达。利用免疫组化以及HE染色方法检测第6、7、8肋软骨和耳软骨中Ⅱ型胶原蛋白和软骨细胞的含量。结果Ⅱ型胶原蛋白m RNA在第6、7、8肋软骨的表达明显高于耳软骨(9.3±1.2、8.9±0.5、9.7±1.3 vs.0.8±0.6);第6、7、8肋软骨的Ⅱ型胶原蛋白相对灰度值明显高于耳软骨(14 545±398、15 807±736、13 257±802 vs.6 310±514);第6、7、8肋软骨测量的软骨细胞数目明显多于患者耳软骨。结论自体第6、7、8肋软骨Ⅱ型胶原蛋白含量高于患者耳软骨,且软骨细胞也更多,自体第6、7、8肋软骨是耳再造术耳支架材料的最佳选择。
Objective To compare the content of type Ⅱ collagen between the autologous 6th,7th, 8th costal cartilages and auricular cartilages. MethodsBy using florescent real-time quantitative PCR methods to test the level of m RNA of type IIcollagen in the autologous 6th, 7th, 8th costal cartilages and auricular cartilages. By using immunohistochemistry and HE staining methods to test the protein content of type IIcollagen and the amount of chondrocytes in the 6th,7th, 8th costal cartilage and auricular cartilage.Results The type II collagen m RNA's level in 6th, 7th, 8th costal cartilages were significantly higher than that in the auricular cartilage(9.3±1.2,8.9±0.5,9.7±1.3 vs. 0.8±0.6);The type Ⅱ collagen's the relative gray value in 6th, 7th, 8th costal cartilages were significantly higher than that in the auricular cartilage(14 545±398,15807±736,13 257±802 vs. 6 310±514;The measurement of chondrocytes in 6th, 7th, 8th costal cartilages were significantly more than that in the auricular cartilage. Conclusion The amount of chondrocytes and the expression of type Ⅱ collagenin autologous 6th,7th,8th costal cartilages were more than auricular cartilage in patients. The autologous 6th, 7th, 8th costal cartilages were the best choice for auricular scaffold on ear reconstruction.
