响应面法优化富硒荷叶离褶伞菌丝体胞内粗多糖发酵条件 被引量:11

Optimizing fermentation condition of intracellular polysaccharide from selenium-enriched Lyophyllum decastes mycelia by response surface methodology

摘要 为了优化富硒荷叶离褶伞菌丝体胞内粗多糖发酵条件,在单因素试验基础上,考察灭菌条件、培养时间、硒添加量和硒添加时间对富硒荷叶离褶伞菌丝体胞内粗多糖的影响,并通过Box-Benhnken实验设计和响应面分析法确定最优发酵条件。结果表明,在121℃、20min条件下对培养基和Na2Se O3溶液分别灭菌后混合,在发酵至第129h,发酵液中添加4μg/m L Na2Se O3,继续发酵至第244h,发酵产生的富硒荷叶离褶伞菌丝体胞内粗多糖含量为349.8mg/100m L。 One-factors-at-a-time experiments were carried out to investigate the effects of sterilizing condition, incubation duration, selenium addition amount and optimal replenishing time on the yield of crude intracellular polysaccharide of selenium-enrichded Lyophyllum decastes mycelia. The process parameters were optimized using a Box-Behnken experimental design coupled with response surface methodology. Culture medium and sodium arsenate solution were mixed after respective sterilization for 20 minutes under 121℃. It was showed that addition of 4μg/m L Na2 Se O3 in 129 h of incubation and continued extension of incubation duration resulted in high yield of crude intracellular polysaccharide in Lyophyllum decastes mycelia, reaching 349.8mg/100 m L in 244 h of fermentation.
