摘要
目的利用肠外翻模型,研究波棱瓜子总木脂素纳米混悬剂(total lignans of Herpetospermum caudigerum nanosuspension,HTL-NS)在大鼠体外的肠吸收特性,并与波棱瓜子总木脂素混悬剂(total lignans of Herpetospermum caudigerum coarse suspensions,HTL-CS)进行对比。方法以波棱甲素与波棱素在大鼠的肠吸收参数为指标,考察不同质量浓度HTL-NS在大鼠十二指肠、空肠、回肠的吸收状况。结果 HTL-NS中2种成分在各肠段的累计吸收量、吸收速率常数和表观渗透系数随着质量浓度的增加而增加,并且明显高于HTL-CS(P<0.05或P<0.01);波棱甲素与波棱素在空肠吸收最好,而十二指肠与回肠的吸收无显著性差异(P>0.05)。结论 HTL在体内的吸收可能为被动吸收,HTL-NS能有效改善HTL肠吸收。
AIM To investigate the characteristics of rats' intestinal absorption of total lignans of Herpetospermum caudigerum nanosuspension( HTL-NS) and compare the absorption difference between HTL-NS and the total lignans of Herpetospermum caudigerum coarse suspensions( HTL-CS) based on the everted gut model in vitro.METHODS The intestinal absorption parameters of herpetrion and herpetin were the indices for the evaluation of absorption characteristics of HTL-NS in rats' duodenum,jejunum and ileum. RESULTS The overall accumulative absorption,absorption rate constants and apparent absorption coefficients of HTL-NS,rising with the increasing concentration,were markedly higher than those of HTL-CS at each intestinal segment( P〈0. 05 or P〈0. 01).There was existed absorption difference in herpetrion and herpetin in different intestine parts. The best absorption site was found in the jejunum,and no significant difference( P〈0. 05) was observed in the duodenum and ileum.CONCLUSION HTL shows its passive diffusion process,and HTL-NS can effectively improve the intestinal absorption of HTL.
