摘要
目的:研究抗β3肾上腺素能受体抗体对心力衰竭大鼠模型心功能的保护作用。方法:选择雄性SD大鼠作为实验动物,采用异丙肾上腺素腹腔注射的方法建立心力衰竭大鼠模型,IgG组给予IgG注射治疗、β3-AA组给予β3-AA注射治疗。检测大鼠血清中β3-AA含量、左心室射血功能和肥厚情况、心肌组织和血清中中左心室重构分子的含量。结果:(1)β3-AA含量:注射β3-AA后10d、20d、30d、40d和50d时,β3-AA组大鼠血清中β3-AA含量均高于IgG组;(2)心功能指标:IgG组大鼠的LVSP、±dp/dtmax均低于Sham组,LVEDP、LVW/BW高于Sham组;β3-AA组大鼠的LVSP、±dp/dtmax均高于IgG组,LVEDP、LVW/BW低于IgG组;(3)心室重构分子:IgG组大鼠血清中血管紧张素Ⅱ、醛固酮的含量以及心肌组织中I型胶原蛋白、Ⅲ型胶原蛋白、ADMA10、ADAM17的含量高于Sham组;β3-AA组大鼠血清中血管紧张素Ⅱ、醛固酮的含量以及心肌组织中Ⅰ型胶原蛋白、Ⅲ型胶原蛋白、ADMA10、ADAM17的含量低于IgG组。结论:抗β3肾上腺素能受体抗体能够改善心力衰竭大鼠模型的心室射血功能、减轻心室肥厚和重构,具有明确的心功能保护作用。
Objective To study the protective effect of antibody against β3-adrenergic receptor on heart function of rat model with heart failure. Methods: Male SD rats were selected as experimental animals. Intraperitoneal injection of isoprotere- nol was used to establish rat model with heart failure. IgG group received IgG injection therapy and β3-AA group received β3- AA injection therapy. Then serum β3-AA content, left ventricular ejection function and hypertrophy, and contents of left ven- tricular remodeling molecules in myocardial tissue and serum of rats were detected. Results= (1) 133-AA content= 10 d, 20 d, 30 d, 40 d and 50 d after β3-AA injection, serum 133-AA contents of 133-AA group were higher than those of IgG group; (2) heart function indexes LVSP and +dp/dtmax of IgG group were lower than those of Sham group, and LVEDP and LVW/BW were higher than those of Sham group; LVSP and ±dp/dtmax of 133-AA group were higher than those of IgG group, and LV- EDP and LVW/BW were lower than those of IgG group; (3) ventricular remodeling molecules= angiotensin II and aldosterone contents in serum as well as type I collagen, type III collagen, ADMA10 and ADAM17 contents in myocardial tissue of IgG group were higher than those of Sham group; angiotensin II and aldosterone contents in serum as well as type I collagen, type III collagen, ADMA10 and ADAM17 contents in myocardial tissue of β3-AA group were lower than those of IgG group. Gon- clusion: Antibody against 133-adrenergic receptor can improve ventricular ejection function of rat model with heart failure and re- duce ventricular hypertrophy and remodeling, and have significant protective effect on heart function.
