摘要
为了研究14种冰草(Agropyron cristatum)在20%聚乙二醇(PEG)模拟干旱胁迫下同工酶的变化,试验采用垂直聚丙烯酰胺凝胶电泳方法对冰草叶片过氧化物同工酶(POD)、过氧化氢同工酶(CAT)和酯酶(EST)同工酶活性的变化进行分析。结果表明:PEG胁迫下14种冰草POD、CAT、EST同工酶酶带数和活性、带级与对照各不相同,POD同工酶谱带共出现181条,最多的有9条,最少的有4条,出现的酶带类型为17种;CAT同工酶酶谱共出现185条,最多的有10条,最少的有4条,出现的酶带类型为15种;EST同工酶酶谱共出现176条,最多的有10条,最少的有4条,出现的酶带类型为16种;3种同工酶的变化与冰草对PEG胁迫的抗性不同有关,蒙农杂种冰草的抗旱性比较强,而中间冰草的抗旱性比较弱。
To study the changes in the isozymes from 14 kinds of Agropyron cristatum under 20% polyethylene glycol (PEG) stress for stimula- ting drought, a method of polyacrylamide gel electmphoresis was used to analyze the changes in the activities of peroxidase ( POD ), catalase (CAT) and esterase (EST) isozymes from 14 kinds of Agropyron cristatum. The results showed that the number of bands, activities, and the band levels of POD, CAT, EST isozymes from 14 kinds ofAgropyron cristatum were different compared with the control under PEG stress. A to- tal of POD isozyme bands appeared 181 bands with maximum and minimum of 9 and 4, and the type of the POD isozyme bands was 17. A total of the CAT isozyme patterns appeared 185 bands with maximum and minimum of 10 and 4, and the type of the CAT isozyme hands was 15. A total of the EST isozyme patterns appear 176 bands with maximum and minimum of 10 and 4, and the type of the EST isozyme bands was 16. The changes in the three kinds of isozymes were related to the difference in the resistance of AgroFyron cristatum to PEG stress. Mengnong hy- brid Agropyron cristatum had strong drought resistance, but intermediate Agropyron cristatum is relatively weak.
