摘要
以陕西茯砖茶中优势菌群冠突散囊菌为试验菌种,采用液体发酵培养,利用石油醚提取色素并对其理化性质进行研究。结果表明,冠突散囊菌黄色素的最大吸收波长为398 nm,该色素不耐高温,60℃以下,色素降解率低于10%,超过60℃,稳定性下降,100℃条件下,保温6h色素降解率为32.87%;太阳光照射7d色素降解率达到28.98%,紫外灯照射7d色素降解率为20.02%pH 2-8条件下色素降解率在8%以下,pH10和12时降解率分别为37.65%和45.98%,该色素耐酸不耐碱;金属离子Fe^(3+)、Fe^(2+)、Cu^(2+)对冠突散囊菌色素有破坏作用,放置48 h后,色素的降解率分别为27.53%、17.89%和38.46%其他金属离子则无不良影响;蔗糖、葡萄糖浓度对色素无影响而VC有护色作用;苯甲酸钠、无机酸和双氧水均有破坏性,放置48 h,色素降解率均超过20%。
Taking Eurotium cristatum,the superiority strains in Fuzhuan brick tea of Shaanxi,as test subjects,use liquid fermentation culture and adopting petroleum ether to extract the pigment then study the physical and chemical properties of it.The results showed that pigment's maximum absorption wavelength was 398 nm;the yellow pigment did not tolerate high temperature and the degradation rate of the pigment was 32.87%after high-temperature treatment for 7 d at 100℃ and declined as the temperature reduces.Light,especially sunlight,showed in an obvious loss of stability of the pigment.Better stability of the pigments was observed at the range pH 2~8.Meanwhile,degradation rate was more than 37.65% at pH 10 and 45.98%at pH 12.Most metal ions had no effects on the stability of pigment except that Fe^(3+),Fe^(2+),Cu^(2+) had severe undesirable effect after 48 h degradation rate was 27.53%,17.89%and 38.46%respectively.The different concentrations of sugar and the high concentration of VC fortified the tone of pigment,while sodium benzoate,hydrogen perodide and inorganic acid obviously degraded the tone of pigment after 48 h,and degradation rate was both more than 20%.
