摘要
[目的]优选老鹰茶总黄酮颗粒剂的制备工艺并建立质量控制方法。[方法]将老鹰茶在一定提取方法下得到其水溶液,再通过大孔树脂纯化得到总黄酮干浸膏粉,通过考察不同辅料对颗粒剂成型性、堆密度、休止角、吸湿性的影响筛选出制备颗粒剂的最优辅料和处方组成。采用薄层色谱法对老鹰茶总黄酮进行定性,通过分光光度法对老鹰茶总黄酮颗粒剂含量进行测定及方法学考察。[结果]老鹰茶总黄酮颗粒剂最佳处方为总黄酮粉末30 g,甘露醇20 g,乳糖10 g;最佳工艺条件为原辅料比1∶1.0 g/g,润湿剂70%乙醇,烘干温度60℃。总黄酮的标准曲线为A=22.100c+0.001 0(R2=0.999 86),颗粒剂的水分、溶化性、粒度均符合要求,总黄酮含量为134.4mg/g,加样回收率96.94%。[结论]老鹰茶总黄酮颗粒剂制备工艺简单、可行,质量稳定可控。
[ Objective ] To study the preparation technology of total flavonoids from Laoying tea granules and to establish methods for its quality control. [ Method] Water solution of Laoying tea was obtained under a given extraction method. Extract powder of total ilavonoids was obtained by macroporous resin. Effects of auxiliary materials on the compactibility, hygroscopicity, bulk density and slope of repose were investigated, so as to screen the optimal auxiliary materials and prescriptions. Total flavonoids from Laoying tea granules were qualitatively analyzed by thin layer chromatography. Content detection and methodology study of total flavonoids were carried out by spectrophotometric method. [ Result ] The opti- mal prescription of total flavonoids from Laoying tea granules were as follows : 30 g powder of total flavonoids, 20 g mannitol and 10 g lactose. The optimal technology was 1 : 1.0 g/g raw materials to auxiliary materials ratio, 70% ethanol as wetting agent and 60℃ baking temperature. The standard curve of total flavonoids was A =22. 100c +0. 001 0(R2 =0. 999 86). The dissolubility, moisture content and grain size all met the requirements. Content of total flavonoids was 134.4 mg/g, the added recovery rate was 96.94%. [ Conclusion ] This technology is simple and feasible; and the quality was stable and controllable.
