江西铅山红芽芋再生苗遗传稳定性的RAPD分析和流式细胞术检测 被引量:4

摘要 通过RAPD分子标记和流式细胞术来分析和检测江西铅山红芽芋再生苗的遗传稳定性,为其种质资源的离体保存和试管苗的工厂化生产提供理论依据。结果表明,经流式细胞术检测,江西铅山红芽芋9种再生苗与对照组的第1个主峰对应的荧光强度值和第2个峰所对应的荧光强度无显著变化。通过20条随机引物对江西铅山红芽芋9种再生苗与对照组进行RAPD扩增。扩增出来的片段大小为200~3 000 bp,20条引物的扩增位点数分别为12,10,7,8,9,14,9,14,10,8,10,10,10,10,11,8,9,10,8,15,共获得202个位点,平均每个引物获得10.1个位点,其中160个为可重复的多态性位点,平均每个引物检测到的多态性位点数为8个,平均多态性位点百分率为79.21%。用NTsys2.10软件对9种再生苗与对照组进行聚类分析,9种再生苗与对照组分成了两类,一类只有1种,即愈伤组织再生苗,另一类包括8种再生苗和对照组。表明愈伤组织再生苗与其他8种再生苗和对照组比较具有显著差异,而其他8种再生苗和对照组之间差异不显著。 The RAPD analysis and flow eytometry detection of genetic stability of Jiangxi Yanshan red bud taro regen- erated planflets were studied, which provide a theoretical basis for conservation in vitro of its germplasm resource and factory production of plantlets. The experimental results showed that, the fluorescence intensity value of the first and the second peak of 9 kinds of regenerated plantlets and the control group of Jiangxi Yanshan red bud taro had no sig- nificant change by flow cytometry detection. DNA of 9 kinds of regenerated plantlets and the control group of Jiangxi Yanshan red bud taro were amplified by RAPD using 20 random primers. The amplified fragment size was 200 - 3 000 bp. The amplification number of 20 primers was 12, 10, 7, 8, 9, 14, 9, 14, 10, 8, 10, 10, 10, 10, 11, 8, 9, 10, 8, 15. 202 site(strip) were obtained, each primer produced 10. 1 site, of which polymorphic loci was160, the average number detected by each primer was 8 and the average percentage of polymorphic loci was 79. 21%. Using NTsys2. 10 software, clustering analysis of 9 kinds of regenerated plantlets and the control group of Jiangxi Yanshan red bud taro was studied, which were divided into two categories, one category had 1 kind, namely callus regenerated plantlets, another category included 8 kinds of regenerated plantlets and the control group. The re- sults also showed that callus regenerated plantlets had significant differences compared with other 8 kinds of regenera- ted plantlets and the control group, while there was no difference between the other 8 regeneration plantlets and the control group.
