摘要
运用荧光光谱、同步荧光光谱、紫外吸收光谱探讨了Co^(2+)对槲皮素(Quercetin,Que)与牛血清白蛋白(Bovine Serum Albumin,BSA)结合作用的影响,初步分析了影响二者结合作用减弱的因素.Co^(2+)的存在使Que与BSA作用的猝灭常数、结合常数、结合位点数均减小,但Co^(2+)没有改变二者的猝灭机制,仍为静态猝灭.Co^(2+)与Que的竞争作用及BSA-Co和Que-Co复合物的形成,抑制了Que与BSA的结合.
The effect of Co2+ on the combination between quercetin(Que)and bovine serum albumin(BSA)has been investigated by the fluorescence spectrum.The weakening factors of the combination between Que and BSA were analyzed preliminarily in the presence of Co2+.The result indicates that the quenching constant(KSV),binding constant(KA)and binding sites(n)all decreased,but the quenching process of BSA was not changed and the quenching mechanism was still a static quenching process when the appropriate amount of Co2+ was added to the mixture of BSA and Que.The anslysis shows that the Co2+ competition role,the BSA-Co complex and Que-Co compound restrained the combination between Que and BSA in Tris-HCl buffer solution.
