姜黄素对血管紧张素Ⅱ刺激下内皮细胞的保护作用与机制研究 被引量:2

Protective Effect and Mechanism of Curcumin on Endothelial Cells Stimulated by Angiotensin Ⅱ

摘要 目的研究姜黄素对血管紧张素Ⅱ刺激下人脐静脉血管内皮细胞(HUVECs)的保护性作用及其机制。方法采用MTT法检测HUVECs的细胞活力;生化比色法和ELISA法检测细胞培养上清中超氧化物歧化酶(SOD)、丙二醛(MDA)、乳酸脱氢酶(LDH)、一氧化氮(NO)及内皮素-1(ET-1)的含量;实时RT-PCR检测HUVECs的Caspase 3、Caspase 9、Bcl-2、Bax基因的mRNA相对表达水平。结果 1×10^(-7)mol·L^(-1)AngⅡ刺激可以使HUVECs活力降低,使细胞培养上清中SOD、NO含量减少,使MDA、LDH及ET-1含量升高。AngⅡ刺激下HUVECs的Caspase 3、Caspase 9、Bax基因的mRNA相对表达水平均有所上调,Bcl-2基因的mRNA相对表达水平有所下调。不同剂量的姜黄素能够减少AngⅡ刺激下对HUVECs的损伤作用,能够上调SOD、NO的含量及Bcl-2基因的mRNA相对表达水平,同时下调MDA、ET-1及LDH含量及Caspase 3、Caspase 9、Bax基因的mRNA相对表达水平。结论姜黄素对于AngⅡ刺激下的HUVECs具有保护作用,其机制可能与减少细胞氧化应激损伤以及抑制细胞线粒体途径凋亡有关。 Objective To investigate the protective effect and mechanism of curcumin on human umbilical vascular endothelial cells(HUVECs) under the stimulation of angiotensin Ⅱ(AngⅡ).Methods MTT assay was used to detect the vitality of HUVECs.Biochemical colorimetric method and ELISA were used to detect the contents of superoxidase dismutase(SOD),malondialdehyde(MDA),nitric oxide(NO),endothelin 1(ET-1) and lactate dehydrogenase(LDH)of the cell culture supernate.And real- time RT- PCR was used to detect the mRNA expression of Caspase 3,Caspase9,Bcl-2 and Bax.Results Stimulation with Angll at 1×10^(-7) mol·L^(-1) reduced the vitality of HUVECs and the production of SOD and NO,upregulated the contents of MDA,ET^(-1) and LDH.Meanwhile,mRNA expression of Caspase 3,Caspase 9 and Bax was upregulated and Bcl-2 mRNA expression was downregulated.Curcumin could eliminate the damage of HUVECs caused by Angll,had an effect on upregulating the contents of SOD,NO and Bcl- 2mRNA expression,and on downregulating the production of MDA,ET-1,LDH and mRNA expression of Caspase 3,Caspase 9 and Bax.Conclusion Curcurrin has protective effects on HUVECs under the sitimulation of Angll,and the possible mechanism is associated with the reduction of oxidative stress damage and with the inhibition of mitochondrial apoptosis.
