当归挥发油分子蒸馏馏分GC-MS分析及抗氧化研究 被引量:13

GC-MS Analysis and Antioxidant Activity Study of Distillates Separated from Essential Oil of Angelicae Sinensis Radix by Molecular Distillation

摘要 目的研究当归挥发油分子蒸馏后各馏分的化学组成及抗氧化能力。方法用分子蒸馏设备对当归挥发油进行分馏,采用气相色谱-质谱法(GC-MS)表征各馏分的化学组成,采用主成分分析法(PCA)分析其化学组成的差异,用2,2-二苯基-1-苦基苯肼(DPPH)自由基清除能力评价各馏分的抗氧化能力。结果当归挥发油经分子蒸馏后得到6个馏分。GC-MS结果显示藁本内酯是各馏分的主要成分。PCA结果显示各馏分除了在Z-藁本内酯上的差异外,馏分6中的油酸和馏分5中的Z-氧代环十七碳-8-烯-2-酮与其他馏分有较大差异。抗氧化能力测定结果显示各馏分都有抗氧化能力但有差异,而且对馏分中藁本内酯的含量无依赖性,馏分6和馏分5的抗氧化能力较强,可能与其所含的特异性成分有关。结论本研究可为当归和其他中药挥发油的进一步开发利用提供参考。 Objective To study composition and antioxidant capacity of distillates separated from the essential oil of Angelicae Sinensis Radix by molecular distillation. Methods The distillates of the essential oil of Angelicae Sinensis Radix were obtained by molecular distillation equipment, and the composition of distillates was analyzed by GC-MS. The principal component analysis(PCA) was used to analyze the difference of the composition of each fraction. The DPPH free radical scavenging ability was applied to evaluate the antioxidant capacity of distillates. Results Six fractions were obtained by molecular distillation from the essential oil of Angelicae Sinensis Radix. GC-MS results showed that ligustilide was the main component in each fraction; PCA results showed that oleic acid in fraction 6 and Z-Oxacycloheptadec-8-en-2-one in infraction 5 were different from the other fractions; The antioxidant capacity results showed each fraction had antioxidant capacity, but they were different, and the capacity of antioxidant were uncorrelated with the content of ligustilide in distillates; antioxidant ability of fraction 6 and fraction 5 was strong, which may be related to specific components. Conclusion This study provided reference for further development of the volatile oil in Angelicae Sinensis Radix and other herbal medicine.
