摘要
目的评估线性探针技术快速检测耐多药结核分枝杆菌的效果。方法选取317例结核分枝杆菌分离株分别进行MGIT960液体药物敏感性试验和线性探针快速检测,以液体法作为金标准,探讨线性探针检测技术的敏感性和特异性以及Kappa值。结果 317株结核分枝杆菌复合群样本分别进行MGIT960液体药物敏感性试验和线性探针快速检测,其中液体法利福平(RFP)耐药率为5.5%,异烟肼耐药率为11.5%,耐多药(MDR)患者检出率为4.7%;线性探针快速检测法利福平(RFP)耐药率为7.4%,异烟肼耐药率为11.7%,MDR检出率为5.7%。以液体法为金标准,线性探针检测技术检测利福平和异烟肼的敏感性和特异性分别为94.3%、97.7%和86.3%、98.0%。结论线性探针检测技术是一个敏感、特异和快速的诊断M DR的有效方法。
Objective To evaluate the application of linear probe technology for rapid detection of multidrug-resistant Mycobacterium tuberculosis( M DR-TB) isolates. Methods Three hundred seventeen Mycobacterium tuberculosis isolates were tested for drug susceptibility by using M GIT 960 liquid method and linear probes technique. With liquid method as gold standard,the sensitivity,specificity and the Kappa value of linear probing method for detection of M DR-TB was evaluated. Result Both M GIT 960 drug susceptibility tests and linear probe detection were performed for a total of 317 Mycobacterium tuberculosis isolates. The M GIT 960 tests showed that the resistant rate to Rifampicin was 5. 5%,Isoniazid was 11. 5% and the rate of M DR-TB was 4. 7%; while linear probe detection showed the resistance rate to Rifampicin was 7. 4%,to Isoniazid was 11. 7% and the rate of M DR-TB was 5. 7%. With M GIT 960 method as golden standard,the sensitivities of linear probe method for detection of resistance to Rifampicin and Isoniazid were 94. 3%,97. 7% and the specifici-ties were86. 3% and 98%,respectively. Conclusion Linear probe technique is a rapid and effective method for detection of M DR-TB.
