摘要
为探究苦参碱在抗炎方面的作用,本试验用LPS建立奶牛子宫内膜上皮细胞炎症损伤模型,通过MTT法测得LPS最佳刺激浓度为30μg/mL,作用12h;苦参碱的低中高浓度分别为5、10、20μg/mL。RT-PCR分别检测3、6、12h各组细胞因子(TNF-α、IL-1β、IL-6、IL-8)mRNA的表达变化,结果表明,苦参碱显著(P<0.05)降低TNF-α、IL-1β、IL-6、IL-8的表达,具有较强的抗炎作用。
The aim of this study was to explore the role of matrine in anti-inflammation.The LPS-induced bovine endometrial epithelial cell model was established.Using MTT assay,LPS optimal stimulation concentration was 30μg/mL for 12hours;the concentration of matrine were 5,10,20μg/mL.RT-PCR was used to detect cytokines(TNF-α,IL-1β,IL-6,IL-8)mRNA expression at 3,6,12 hour.The results showed that matrine significantly reduced TNF-α,IL-1β,IL-6,IL-8 mRNA expression.Matrine shows a strong anti-inflammation effect.
