摘要
目的观察肾上腺素(Adr)对新生大鼠心肌细胞体积的影响,并探讨其机制。方法原代培养新生大鼠心肌细胞,分为观察组和对照组,观察组加入Adr(1μmol/L),对照组加入等体积PBS,培养48 h。采用Count Star自动细胞计数仪测量各组心肌细胞体积变化;荧光显微镜观察心肌细胞氯通道蛋白3(Cl C-3)荧光强度,在所有拍摄条件相同尤其是曝光时间相同的条件下对心肌细胞拍照,用Image Pro Plus分析软件定量分析心肌细胞Cl C-3的平均光密度(AOD);Western blotting法检测心肌细胞Cl C-3。结果观察组及对照组心肌细胞体积分别为(964.80±60.00)、(716.28±57.49)μm3,心肌细胞Cl C-3的AOD分别为0.037±0.012、0.062±0.009,心肌细胞Cl C-3的相对表达量分别为0.372±0.027、1.040±0.210,两组比较,P均<0.05。结论 Adr能诱导新生大鼠心肌细胞肥大,其机制可能与下调Cl C-3表达有关。
Objective To observe the effect of adrenaline (Adr) on volumes of cardiomyocytes in neonatal rats and to investigate its mechanism. Methods The cardiomyocytes of neonatal rats were cultured in vitro and were divided into the observation group which was added with 1 μmol/L Adr for 48 h and the control group which was added with the same vol- umes of PBS for 48 h. The volume of cells were measured by CountStar. The fluorescence intensity of chloride channel pro- tein 3 (C1C-3) protein was detected by immunofluorescence. The average optical density (AOD) of C1C-3 was measured by Image Pro Plus or Image J software. Western blotting was used to detect C1C-3. Results The volume of cardiomyocytes in the observation group and the control group was (964.80 ±60.00) and (716.28 ±57.49) μm3 respectively. The AOD in the two groups was 0. 037 ± 0. 012 and 0. 062 ± 0. 009. The C1C-3 protein expression in the two groups was 0. 372 ± 0.027 and 1. 040 ± 0. 210. Significant difference was found between these two groups ( all P 〈 0.05 ). Conclusion Adr can induce the cardiac hypertrophy of neonatal rats, and the mechanism may be related with the down-regulated expressionof C1C-3.
