Bax/Bcl-2基因在成年辽宁绒山羊皮肤次级毛囊表达及年周期变化规律 被引量:3

摘要 绒山羊的皮肤毛囊具有在一个生物年内呈周期性发育的特性,已有研究表明,在绒山羊毛囊周期性发育的各时期存在细胞凋亡现象。而作为调控细胞凋亡的Bax/Bcl-2基因在绒山羊皮肤毛囊的表达是否与其各个发育时期相关,尤其是二者在毛囊发育哪个时期为优势基因,将对阐明绒山羊毛囊发育过程中细胞凋亡所发挥的生物机制具有重要意义。本研究采用免疫组化方法,检测成年辽宁绒山羊母羊皮肤中Bax和Bcl-2蛋白表达部位及表达量年周期变化情况。研究结果表明:在不同月份辽宁绒山羊成年母羊皮肤次级毛囊的毛球、内根鞘、外根鞘上,以及汗腺、皮脂腺以及表皮上均可见Bax和Bcl-2阳性表达;次级毛囊Bax表达量在5月份和9月份最高,Bcl-2表达量在5月份最高,次级毛囊Bax/Bcl-2比值在每年的4月份和7月份出现2个峰值,10月则为全年的最低点。 The hair follicle of cashmere goat has the characteristic of periodic development in a biological year. The research showed that there exist cell apoptosis in the different periods of the periodic development of the hair follicle. The expression of Bax/Bcl-2 gene in cashmere goat skin is related to the development of the hair follicle of cashmere goat, especially in the development of hair follicle. The dominant gene will be of great significance to the biological mechanism of cell apoptosis in hair follicle development: In this research,the expression of Bax and Bcl-2 and the distribution of micro vessel was detected by using immunohistochemisty. The results showed as follow,by studying the adult female Liaoning Cashmere Goat monthly in a whole year,Bax and Bcl-2-positive were detected in PF and SF hair bulb,inner root sheath, outer root sheath, and aebaceous gland, sweat gland,pidermis,similar to VEGF. Expression of Bax in PF minimized at June and maximized at July,while Bcl-2 was unremarkable among months,thus the ratio of Bax/Bcl-2 in PF minimized at June and maximized at July. Expression of Bax in SF was bimodal pattern with two peaks at May and September,while an only peak of Bcl-2 shown at May,thus the ratio of Bax/Bcl-2 in SF minimized at October and maximized
