食管鳞状细胞癌组织中DMRT2的表达及临床意义

摘要 目的检测食管鳞状细胞癌(esophageal squamous cell carcinoma,ESCC)组织中的DMRT2(double-sex and mab-3 relatated transcription factor 2)蛋白及m RNA表达情况,并分析其与临床病理资料的关系。方法制作微阵列组织芯片,利用免疫组织化学DAB法检测64对ESCC(实验组)及癌旁正常食管黏膜组织(对照组)中DMRT2蛋白的表达情况,并统计分析DMRT2蛋白表达水平与临床病理资料的关系。利用QRT-PCR技术检测实验组及对照组中DMRT2 m RNA表达情况。结果 (1)免疫组织化学结果显示ESCC组织中DMRT2蛋白表达明显低于癌旁正常黏膜组织(χ2=17.926,P=0.000)。(2)DMRT2蛋白低表达与淋巴结转移有关(P=0.026),而与性别、大体分型、分化、分期无关。(3)QRT-PCR结果显示m RNA在食管鳞癌组织中表达量明显低于正常黏膜组织(t=7.108,P=0.000)。结论DMRT2低表达可能与食管鳞状细胞癌的发生发展有关,并可能参与肿瘤的转移复发过程。 Objective To investigate DMRT2 protein and mRNA expression in esophageal squamous cell carcinoma (ESCC) tissues, and analyze their relationships with clinicopathological data. Methods Using tissue microarray and immunohistochemistry (IHC), the expression of DMRT2 were determined in 64 pairs ESCC tissues(observation group) and their corresponding normal esophageal tissues(control group), and we analyzed its correlation to the clinicopathological characteristics. Besides, we used QRT-PCR to detect DMRT2 mRNA expression in the observation group and control group. Results (1) The expression of DMRT2 protein in ESCC tissues was significantly lower than that in paraneoplasfic normal esophageal tissues. (49.1% vs. 89.5%, Z2=17.926, P〈0.001). (2) Low DMRT2 protein expression was associated with lymph node metastasis (P=0.026), but not associated with gender, tumor cell differentiation, general classification or stages. (3) QRT-PCR showed that DMRT2 mRNA expression in ESCC tissues was significantly lower than that in paraneoplastic normal esophageal tissues. Conclusion Low DMRT2 expression may be involved in the occurrence and development of ESCC, and may be correlated to tumour metastasis and recurrence.
