摘要
目的:观察佛手黄酮对高脂血症兔血脂及动脉粥样硬化相关危险因子的作用及机制。方法:28只新西兰兔随机分为四组:对照组、模型组、佛手黄酮组和氟伐他汀组,每组各7只。对照组给予基础饲料喂养,模型组高脂饲料喂养,佛手黄酮组及氟伐他汀组在高脂饲料喂养基础上,分别给予佛手黄酮100mg/kg/天灌胃及氟伐他汀10mg/kg/天灌胃。10周后采用ELISA等测量各组血清总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)、一氧化氮(NO)、丙二醛(MDA)和白细胞介素-1β(IL-1β)水平,采用Western blotting检测各组肝组织载脂蛋白E(ApoE)表达水平。结果:与对照组比较,模型组TC、LDL-C显著升高(P<0.01),佛手黄酮组TC、LDL-C、MDA、IL-1β水平较模型组显著降低(P<0.05),NO和肝组织AopE蛋白表达较模型组显著升高(P<0.05);效果与氟伐他汀组相当(P>0.05)。结论:佛手黄酮具有调节血脂和抗动脉粥样硬化作用。
Objective:To explore the effect and mechanism of total flavonoids from bergamot on hyperlipidemia and atherosclerosis in rabbits.Method:28rabbits were randomly divided into four groups:control group,model group,total flavonoids from bergamot group and fluvastatin group,every group was 7cases.Rabbits in control group were fed with nomal food,the other three groups were fed with high-fat food,and rabbits in the total flavonoids from bergamot and fluvastatin group were intragastricaly administrated with corresponding drug at dose of100mg/kg and 10mg/kg daily,respectively.After 10 weeks,serum TC,TG,LDL-C,HDL-C,NO,MDA and IL-1βwere deteaed,as well as aopE in liver was tested by Western blotting.Results:Compared with the control group,the levels of TC and LDL-C in model group were significantly increased(P〈0.05).Compared with the model group,the levels of TC,LDL-C、MDA and IL-1βin total flavonoids from bergamot group were significantly decreased(P〈0.05),while the NO and the expression of AopE protein in liver tissue were significantly increased(P〈0.05),the effect of this group was comparable with the fluvastatin group(P〉0.05).Conclusion:Total flavonoids from bergamot can inhibit hyperlipidemia and atherosclerosis.
