摘要
以如意草为原料,在冷浸预处理基础上,分别采用超声波及热回流法对总生物碱提取得率进行研究。通过正交试验研究乙醇浓度、料液比、提取温度、提取时间对提取效果的影响,确定超声波与热回流提取如意草总生物碱的最佳提取条件,并比较最佳提取条件下两种提取方法的总生物碱得率。结果表明,冷浸-超声波提取最佳条件为提取温度40℃,提取时间150min,料液比1∶10、乙醇浓度70%;冷浸-热回流提取最佳条件为回流时间180min,液料比1∶25,回流温度55℃,乙醇浓度85%。超声波提取较回流法提取具有提取率高,提取时间短、提取温度低、乙醇浓度低等优点。
Corydalis taliensis Fr. was used as material, the alkaloid was extracted through refluxing method and ultrasonic method assisted by cold-soaked pretreatment. The yield of alkaloid as the evaluation index, the concentration of ethanol, solid-to-liquid ratio, extracting time and extracting temperature were optimized by orthogonal test. The results showed that the optimum conditions for ultrasonic method were : temperature 40℃, extracting time 150min, solid-to-liquid ratio 1 : 10 and the ethanol concentration 70%. The optimum conditions for refluxing method were : extracting time 180min, solid-to-liquid ratio 1 : 25, temperature 55℃ and the ethanol concentration 85%. In contrast with refluxing method, ultrasonic extraction has the advantages of high extraction rate, short extraction time, low temperature and low concentration of ethanol.
