摘要
目的:探讨丹皮酚对Si Ha细胞增殖及COX-2转录的影响。方法:以人宫颈鳞癌Si Ha细胞系作为体外模型,MTT法检测丹皮酚对Si Ha细胞增殖的影响。用COX-2启动子质粒和AP-1结合位点质粒转染Si Ha细胞,采用双荧光素酶报告基因检测系统检测COX-2的转录水平和AP-1的活性。结果:MTT实验结果显示丹皮酚显著抑制Si Ha细胞的增殖(P<0.01),且与丹皮酚剂量及干预时间呈正相关。荧光素酶检测结果显示丹皮酚显著降低Si Ha细胞COX-2的转录水平(P<0.01),同时显著抑制AP-1的活性。结论:丹皮酚可通过抑制转录因子AP-1的活性,进而抑制COX-2的转录,从而抑制Si Ha细胞的增殖。
Objective: To investigate the effect of Paeonol on the proliferation and COX- 2 transcription of Si Ha cells.Methods: Human cervical squamous cell carcinoma Si Ha cell line was used as a model in vitro. The proliferation of Si Ha cells treated by Paeonol was detected by MTT assays. Plasmids p COX2- Luc and p AP1- Luc were transfected into Si Ha cells and the dual luciferase reporter gene assay system was used to detect the transcription level of COX- 2 and the activity of AP- 1. Results: The results of MTT assays showed that Paeonol significantly inhibited the proliferation of Si Ha cells and the effect was associated positively with the dose and treatment time of paeonol. Luciferase assay results showed that Paeonol apparently reduced the transcription level of Si Ha COX- 2 cells( P 0. 01),and obviously inhibited the activity of AP- 1( P 0. 01). Conclusion: Paeonol may inhibit COX- 2 transcription by depressing the activity of transcription factor AP- 1,thereby suppress the proliferation of Si Ha cells.
