高效液相色谱法同时测定复方积雪草片中的羟基积雪草苷、积雪草苷、桦木酸和迷迭香酸的含量 被引量:3

Simultaneous determination of madecassoside, asiatiocside, betulinic acid and rosmarinic acid in Fufang Jixuecao tablets by HPLC

摘要 目的建立HPLC法同时测定复方积雪草片中羟基积雪草苷、积雪草苷、桦木酸和迷迭香酸含量的方法。方法采用依利特C_(18)色谱柱,流动相为乙腈(A)-0.1%磷酸溶液(B),梯度洗脱;流速为0.9 m L·min^(-1);柱温为25℃;进样量20μL;羟基积雪草苷和积雪草苷检测用波长为λ1=205 nm,桦木酸检测用波长为λ2=210 nm,迷迭香酸检测用波长为λ3=325 nm。结果羟基积雪草苷、积雪草苷、桦木酸和迷迭香酸在50 min内得到良好的分离,线性范围分别为7.82~156.40μg·m L^(-1)(r=0.9994)、5.95~119.00μg·m L^(-1)(r=0.9992)、6.15~123.00μg·m L^(-1)(r=0.9991)、4.70~94.00μg·m L^(-1)(r=0.9997);4种成分的平均加样回收率(n=6)分别为99.1%、96.9%、98.1%、96.9%,RSD分别为1.4%、1.0%、0.93%、0.86%。结论本文建立的方法可同时测定复方积雪草片中羟基积雪草苷、积雪草苷、桦木酸和迷迭香酸含量,精密度、稳定性和重复性良好。 Objective To establish an HPLC method for simultaneous determination of madecassoside, asiatioc- side, betulinic acid and rosmarinic acid in Fufang Jixuecao tablets. Methods Hypersil C18 column was used, with a mobile phase consisting of acetonitrile-0.1% phosphoric acid solution in a gradient elution. The flow rate was 0.9 mL ·min -1. The column temperature was 25 ~C. The injection volume was 20 μL. Madecassoside and asiatiocside were detected at 205 nm, betulinic acid was detected at 210 nm, and rosmarinic acid was detected at 325 nm. Results Madecassoside, asiatiocside, betulinic acid and rosmarinic acid were well separated in 50 min and showed good linearity at 7.82 - 156.40μg ·mL- 1 (r = 0.999 4), 5.95 - 119.00μg ·mL-1(r = 0.9992), 6.15 - 123.00 μg ·ml-1 (r = 0.9991), and 4.70 - 94.00 μg · mL- 1 (r = 0.9997). The average recoveries and RSDs were 99.1% (1.4%), 96.9% (1.0%), 98.1% (0.93%), and 96.9% (0.86%), respectively. Conclusion HPLC can simultaneously determine the contents of madecassoside, asiatiocside, betulinic acid and rosmarinic acid in Fufang Jixuecao tablets precisely, stably and repeatably.
