摘要
从土壤和城市生活污水处理厂二沉池污泥中筛选出高效絮凝剂产生菌,并研究培养条件对其产絮活性的影响,为高效絮凝剂产生菌的应用提供了理论依据。通过单因素和正交实验对絮凝剂产生菌菌株培养条件进行优化。结果共筛选出24株细菌,其中有明显絮凝活性的菌株共8株,经过复筛获得1株高效稳定絮凝剂产生菌,命名为MBFS-17,经分子生物学鉴定此菌为芽孢杆菌。实验结果表明:MBFS-17菌株的最适碳源是蔗糖,最适氮源是鱼蛋白胨,在碳源浓度为14 g/L,氮源浓度为0.5 g/L,温度为30℃,pH为8的条件下,MBFS-17菌株所产絮凝剂对高岭土悬浊液的絮凝率高达94.7%。筛选得到的絮凝剂产生菌经过培养基优化具有较高的絮凝效果,在废水絮凝处理中有较大的应用前景。
From soil and urban sewage treatment plant in the second pond sludge filter out high efficient flocculant, culture conditions, the table on the flocculation effect was studied for the application of high efficient flocculant produced bacteria provide a theoretical basis. A strain was screened from soil and sludge, and named MBFS -17 ,which was identified as BaciUus and has the highest flocculating activity. Through the single factor and orthogonal experiment, it optimized the culture conditions and the results showed that sucrose is the best carbon source, fish peptone is the best nitrogen source. When the concen- tration of sucrose and fish peptone is 14 g/L, O. 5 g,/L,temperature is 30 ℃ ,pH was 8 ,the bioflocculant was produced by MBFS - 17 achieved the high flocculating activity which reached to 94. 7%. The floeculant produced by the flocculant has a higher flocculation effect, and has great application prospect in the treatment of wastewater flocculation.
