摘要
分离疑似球虫病病鸡的盲肠,刮取其内容物;经多级筛网滤过,获得含有大量卵囊的混合物;饱和盐水法初步分离卵囊;梯度浓度的蔗糖溶液纯化卵囊;将纯化卵囊置于2.5%的重铬酸钾溶液、28℃条件进行孢子化;将孢子化的卵囊用2%琼脂溶液包囊,用高倍显微镜观察,用刀片分割琼脂球虫块至单个卵囊;接种鸡进行回归实验;分别纯化由单个卵囊裂殖的球虫卵囊;将获取的单卵囊饲喂至7日龄雏鸡,获得了5组球虫卵囊;分别用形态学和分子生物学方法鉴定所获得的5组卵囊均为鸡柔嫩艾美耳球虫卵囊;该卵囊的获得为后续研究中草药对鸡柔嫩艾美耳球虫卵囊作用机理的研究提供了材料基础。
The cecum was excised from chicken with clinical suspected coccidiosis and the the content was collected; serial sieve mesh was used to acquire the mixture with generous oocysts and saturated brine was applied to separate the oocysts from the residue; sucrose with gradient density was utilized to purify the ooeysts and sporulated oocysts was generated at the potassium dichromate solution(with 2.5% concentration)and with the temperature of 28℃ ; agar solution at 2% concentration was used to scatter the oocysts and the blade was used to separate the oocysts one by one with the aid of high power microscope; 7 d-old chicken were used to proliferate the oocysts and 5 group oocysts were aquired; All 5 oocysts had typical characteristic of Eimeria tenella by the identification of morphology and Molecular biology methods; acquirement of purified Eimeria tenella ensures the follow-up studies about the mechanism of action of herbs on homemade chicken Eimeria tenella infection.
