结直肠癌组织RUNX3和MMP-2表达与MVD相关性研究 被引量:6

Expressions and correlation studies of RUNX3,MMP-2 and MVD in colorectal adenocarcinoma tissues

摘要 目的 结直肠癌组织抑癌基因RUNX3(human runt-relatedtranscriptionfactor-3)与基质金属蛋白酶-2(matrix metalloproteinases-2,MMP-2)表达以及与微血管密度(microvessel density,MVD)相关性尚未完全阐明,通过免疫组化检测RUNX3、MMP-2和MVD,分析其相关性,探讨三者之间及其与临床病理因素的关系,为探索结直肠癌发病机制及抗血管浸润治疗提供科学依据。方法 收集河北北方学院附属第一医院2013-07-01-2014-12-01手术切除的结直肠癌组织85例和40例癌旁正常组织,应用免疫组化SP法检测85例结直肠癌组织中RUNX3和MMP-2的表达,以及CD34标记的MVD情况。结果RUNX3在结直肠癌组织中阳性表达率为25.88%(22/85),明显低于癌旁正常组织的77.50%(31/40),χ^220.485,P〈0.01;MMP-2在结直肠癌组织中阳性表达率为72.94%(62/85),明显高于癌旁正常组织的22.50%(9/40),χ^231.964,P〈0.01;结直肠癌组织中MVD表达值为17.25±2.16,明显高于正常组织的5.14±2.58,t=32.00,P〈0.01。RUNX3、MMP-2与肿瘤浆膜浸润、TNM分期、CEA(+)、淋巴结转移及肝转移明显相关;癌组织中RUNX3阳性表达组MVD值为11.26±3.45,明显低于阴性组的18.09±4.20,t=8.51,P〈0.01;而癌组织中MMP-2阳性组MVD值为19.11±3.25,明显高于阴性组的10.21±4.04,t=11.77,P〈0.01;且结直肠癌组织中RUNX3与MMP-2呈明显负相关性,r=-0.365,P〈0.01。结论 RUNX3低表达和MMP-2高表达与结直肠癌的发生、发展、侵袭和转移密切相关,二者的异常表达可能是诱导结直肠癌微血管生成的重要原因。 [-ABSTRACT] OBJECTIVE To explore the expressions of RUNX3 and MMP-2 in colorectal carcinoma tissues and their correlation with microvascular generation and clinieopathologic feature. METHODS From 2013 07 01 to 2014 12 01, 85 cases of eolorectal adenocarcinoma and 40 normal colorectal tissues were collected in the First Hospital Affiliated to Hebei North College. Immunohistochemistry staining SP was used to detect the expression level of RUNX3 and MMP 2. Using CD34 as the specific marker, microvascular density (MVD) in eoloreetal adenoearcinoma and normal colorectal tis sues were calculated. RESULTS The positive rate of RUNX3 was 25.88±±(22/85) and 77.50± (31/40) in colorectal ad enocarcinoma tissues and normal colorectal tissues, there was significant difference(x2 = 20. 485, P±0.01) ; The positive rate of MMP-2 was 72.94± (62/85) and 22.50% (9/40) in colorectal adenocarcinoma tissues and normal coloreetal tis- sues, there was significant difference(X2=31. 964,P〈0.01) ; the MVD value (17.25±2.16) in colorectal adenocarcinoma tissues was significantly higher than those (5.14± 2.58) in normal coloreetal tissues, there was significant difference (t= 32.00,P%0.01); The RUNX3 and MMP 2 had significant relations with the depth of tumor invasion, TNM stage, CEA(+), lymph node metastasis and liver metastasis. The value of MVD with RUNX3 positive expression (11. 26 ± 3.45) was significantly lower than that with RUNX3 negative expression in colorectal adenocarcinoma tissues (18.09 ± 4.20), t= 8.51, P%0.01 ;while the value of MVD with MMP-2 positive expression(19.11 ± 3.25) was significantly higher than that with MMP-2 negative expression in colorectal adenocarcinoma tissues(10.21 ± 4.04), t= 11. 77, P〈0.01. The expression of RUNX3 showed significantly negative correlation with that of MMP-2 in colorectal adenocarcinoma tissues(r= -0. 365,P±0.01). CONCLUSION The low expression of RUNX3 and high expression of MMP-2 have the close relations with the occurrence, development, invasion and metastasis of colorectal cancer and their abnormal expression may be the important reason for microvascular formation of colorectal cancer.
